Dose‐dependent initiation of myogenesis by neurotrophic factor

Abstract: The kinetics of primary chick muscle cells in response to partially purified neurotrophic factor (NTF) were investigated. Chick muscle cells are routinely plated in the absence of NTF but in the presence of horse serum to allow cell attachment to the substratum; a maximum number of cells attaches 5-8 hr after plating, but cells do not proliferate in the absence of NTF, or do so extremely slowly. Subsequent to a routine attachment period of 6 hr, the medium is aspirated and replaced with medium containing 2-20 … Show more

“…Cell culture Primary chick muscle cells were cultured in the presence of NTF as previously described (Popiela and Ellis, 1981;Popiela et al, 1982). Muscle cells were dissociated from 11-day-old chick embryo leg muscle tissue in 0.05% collagenase (Millipore, San Francisco, CA).…”

“…Two types of tissue extracts are generally added to the culture medium for growth of primary chick muscle cells. Traditionally, chick embryo extract is chosen (Konigsberg, 1979), or more recently a n aqueous extract of adult chicken ischiatic-peroneal nerves (NE) (Popiela, 1978), or its purified derivative (NTF) (Popiela and Ellis, 1981;Popiela et al, 1982), is substituted instead. The requirement for chick embryo extract was identified by its stimulatory effects on muscle cell proliferation (Slater, 1976), a requirement that is similarly fulfilled by NE or NTF.…”

“…The active substance in these various preparations has now been shown to be identical, or nearly so, to the circulating iron transport protein, transferrin (Tf) (Ii et al, 1982;Ozawa et al, 1981;Popiela et al, 1982;Beach et al, 1983Beach et al, , 1985. In addition to iron transport, Tf is required for the growth and development of a number of mammalian clonal cell lines and several primary cell culture systems also appear to require Tf Sato, 1979, 1985).…”

“…The importance of the extracellular matrix in skeletal-muscle development has been well established (Hauschka & Konigsberg, 1966;Konigsberg, 1970; Hauschka, 1972;Hauschka & White, 1972;De La Haba et al, 1975;Lipton, 1977;Puri et al, 1980;Chiquet et al, 1981;Ehrisman et al, 1981;Popiela et al, 1982;Beach et al, 1983).…”
