Down-regulation of 14-3-3 zeta sensitizes human glioblastoma cells to apoptosis induction

Abstract: Strong 14-3-3 zeta protein expression plays an important role in tumorigenesis, including in the maintenance of cell growth, resistance increase, and the prevention of apoptosis. In this study, we focus on two targets: (1) the expression of 14-3-3 zeta in the different grades of human astrocytoma (II-IV), (2) suppression of 14-3-3 zeta protein expression in glioblastoma derived astrocytes by 14-3-3 zeta shRNA lentiviral particles. The tissues of human astrocytoma were provided from 30 patients (ten of each gra… Show more

“…Also, the pro-survival regulation of cellular responses may be supported by the regulatory action of CBD causing increased levels of theta protein 14-3-3 [ 65 , 66 ] which are decreased by H 2 O 2 . Literature data also indicate an association between down-regulation of protein 14-3-3 and apoptosis caused by ROS production including hydrogen peroxide [ 67 ]. The pro-survival effect of CBD can also be associated with the prevention of the formation of non-physiological modifications in proteins by reducing the formation of protein adducts with lipid peroxidation products and, consequently, the protein cross-linking and aggregation.…”

“…Parasitic infection induces autophagy and cell apoptosis in insects [6]. Cathepsin B and 14-3-3 zeta both have important roles in apoptosis [38,39]. RNAi-mediated downregulation of cathepsin B or the absence of cathepsin B induces apoptosis in cancer [38,40,41].…”

“…Strong 14-3-3 zeta protein expression acts in cell differentiation, proliferation, transformation, and prevention of apoptosis [42]. In particular, 14-3-3 protein zeta is a major regulator of apoptotic pathways in insects and vertebrates [43,44,45,46]; downregulation of 14-3-3 zeta sensitizes cells to apoptosis [39,47]. Cathepsin B (spots 43) and 14-3-3 zeta (spot 45) were successfully identified and were significantly downregulated in the hemolymph of parasitized female silkworm pupae.…”

“…15,16 Downregulation of 14-3-3ζ sensitizes human glioblastoma cells to apoptosis induction, with higher levels of 14-3-3ζ being associated with a higher grade of glioma. 17 Higher levels of 14-3-3 are also evident in GSC. 18 As indicated above, both miR-7 and miR-451 can repress 14-3-3ζ in both tumour and nonneoplastic cells.…”
