Doxorubicin-Loaded Melanin Particles for Enhanced Chemotherapy in Drug-Resistant Anaplastic Thyroid Cancer Cells

Abstract: Anaplastic thyroid cancer (ATC) is highly aggressive and tends to develop drug resistance to standard chemotherapies. To overcome the drug resistance of ATC, we synthesized dopamine-melanin nanoparticles (MNPs) loaded with doxorubicin (Dox) molecules. The Dox-loaded MNPs (Dox-MNPs) developed exhibited increased cellular uptake and enhanced therapeutic efficacy in drugresistant ATC cell line HTh74R, compared with free Dox. The native MNPs were found to have excellent biocompatibility, which suggests that Dox-MN… Show more

“…Most recent research is focused on the development of nanocarriers which can effectively deliver the therapeutic agents on the target site, without early leakage, and consequently release upon a specific stimuli, such as NIR irradiation also in combination with pH [ 41 ]. This behavior can be achieved by simple or modified MNP [ 151 , 185 ]. Combining drug binding capacity of MNP with their innate PTT efficacy, Zhang et al developed an outstanding synergistic PTT-CT cancer treatment [ 186 ].…”

“…The cellular uptake and therapeutic efficacy were significantly higher than those induced by the same amount of free doxorubicin, reaching a cell viability of 34.6 ± 5.4%, at the highest concentration of 160 mg/L. Moreover, no significant cytotoxicity to normal cells was noticed, proving the biocompatibility of the nanoparticles [ 123 ]. Another research team investigated the properties and activity of polyethylene glycol (PEG)-conjugated melanin nanoparticles loaded with varied concentrations of doxorubicin.…”

“…Currently, there are few nanoplatforms developed for drug-resistant ATC ( Marano et al, 2016 , 2017 ; Wang et al, 2018 ). In our previous research, we constructed Dox-loaded melanin nanoparticles to enhance the efficacy of chemotherapy to drug-resistant ATC ( Wang et al, 2018 ).…”

“…Currently, there are few nanoplatforms developed for drug-resistant ATC ( Marano et al, 2016 , 2017 ; Wang et al, 2018 ). In our previous research, we constructed Dox-loaded melanin nanoparticles to enhance the efficacy of chemotherapy to drug-resistant ATC ( Wang et al, 2018 ). However, the Dox-loading capacity of melanin nanoparticles was less than 20%, which may be related to their limited surface-area-to-volume ratio ( Wang et al, 2018 ).…”
