General biochemical biomarkers are widely used in current ecotoxicology and may function as early warning signals. We have, however, poor knowledge on how ecologically similar species differ in their biomarker responsiveness and how predation risk may affect these biomarkers, potentially in an interactive way with pesticides. We evaluated this by exposing four corixid water bug species to combinations of endosulfan and predation risk and quantifying the activity of four general enzymatic biomarkers: acetylcholinesterase (AChE), phenoloxidase (PO), catalase (CAT) and superoxidedismutase (SOD). AChE activity was inhibited at an endosulfan concentration of 2 μg l(-1) and this did not differ significantly among species. Predation risk inhibited AChE activity with the same magnitude as endosulfan in one species, S. striata. Reduction in the investment of immune function following pesticide exposure, as measured by the activity of PO, was only observed in C. coleoptrata at 8 μg l(-1) while we observed an increase of PO levels in S. striata. Overall, PO was suppressed under predation risk at 8 μg l(-1) endosulfan. For SOD we observed a pesticide-induced increase across all species under predation risk, while for CAT the pesticide-induced increase was only present without predation risk. These results indicate that even within this group of ecologically similar and closely related species opposing biomarker responses may exist, as observed for PO. Effects of predation risk on all four enzymes, at a similar magnitude as the pesticide effects, further question their usefulness as general biomarkers.

The distribution of hexachlorocyclohexanes (HCHs) and dichlorodiphenyltrichloroethanes (DDTs) in the surface seawater and sediment of Jincheng Bay mariculture area were investigated in the present study. The concentration of total HCHs and DDTs ranged from 2.98 to 14.87 ng L −1 and were < 0.032 ng L −1 , respectively, in surface seawater, and ranged from 5.52 to 9.43 and from 4.11 to 6.72 ng g −1 , respectively, in surface sediment. It was deduced from the composition profile of HCH isomers and DDT congeners that HCH residues derived from a mixture of technical-grade HCH and lindane whereas the DDT residues derived from technical-grade DDT and dicofol. Moreover, both HCH and DDT residues may mainly originate from historical inputs. The hazard quotient of α-HCH, β-HCH, γ-HCH and δ-HCH to marine species was 0.030, 0.157, 3.008 and 0.008, respectively. It was estimated that the overall probability of adverse biological effect from HCHs was less than 5%, indicating that its risk to seawater column species was low. The threshold effect concentration exceeding frequency of γ-HCH, p,p '-DDD, p,p'-DDE and p,p'-DDT in sediment ranged from 8.3% to 100%, and the relative concentration of the HCH and DDT mixture exceeded their probable effect level in sediment. These findings indicated that the risk to marine benthos was high and potentially detrimental to the safety of aquatic products, e.g., sea cucumber and benthic shellfish.

A systematic literature search identified publications relevant to repeated (94 papers) and mixture exposure (152 papers) of aquatic organisms. Limited data are available on pulsed exposure of pesticides and reports on repeated pulse or fluctuating exposures are even rarer. Although there has been recognition of the need for more realistic testing scenarios these lack a systematic approach. Among the reports on pesticide mixtures most of the plant studies evaluated interactions between mixtures of herbicides or fungicides / algicides. A small number of reports evaluated the effects on invertebrates of mixtures of pesticides with the same mode of action and these showed additive toxicity. More studies in invertebrates assessed interactions between pesticides with different modes of action including herbicide/ insecticide combinations some of which showed toxicity greater than additive. The majority of studies in fish have investigated the toxicity of insecticide mixtures. The toxicity of most combinations was at most additive, with a small number antagonistic. A small number of papers reported mixture effects of pesticides in amphibians; more than additive effects were observed with atrazine combinations. Simple models can be used to determine the additive toxicity of pesticide mixtures based on concentration addition or independent action. More complex models to determine the scale of synergy are species-and dose-dependent and rely on an understanding of the interactions involved. Further work is required to develop a greater understanding of the species, dose and time dependence of exposure to pesticide mixtures in which more complex interactions occur. In the interim, in the majority of cases the increase in toxicity is no greater than 3 fold when compared to additive, the deviation from additivity decreases with the increasing number of components in the mixture, and where no known interactions have been reported this may be an appropriate factor to include in risk assessments.
