Abstract: Benorylate, an esterification product of paracetamol and acetylsalicylic acid is indicated in the treatment of rheumatoid and osteo-arthritis, other painful conditions and febrile states. In animals benorylate’ has been shown to be of low toxicity and only has teratogenic activity at high doses. Gastric effects are less severe than occur with aspirin. “…First ester, – 4-acetamido-phenyl-2-acetoxybenzoate 46 , is known as benorylate drug (Benoral®, Benortan®). 112,113 Its initial hydrolysis of the acetyl group yields a minor metabolite ester 47 , known as acetaminosalol. Its ester bond is enzymatically degradable in the body to produce two active metabolites, acetylsalicylic acid 9 or APAP.…”

“…Clinically, the preparation is used in a pure form in allergic rhinitis, where it diminishes nasal obstruction by virtue of its vasoconstrictor effect and gives good symptomatic relief. It has given equally good effect when administered orally or locally in the nose (3,4,18,23). Both compounds, brompheniramine and phenylpropanolamine, are released successively from the Lunerin tablet, which forms a porous plastic matrix.…”
