Dual function of an Arabidopsis transcription factor DREB2A in water-stress-responsive and heat-stress-responsive gene expression

Abstract: Transcription factor DREB2A interacts with a cis-acting dehydration-responsive element (DRE) sequence and activates expression of downstream genes involved in drought-and salt-stress response in Arabidopsis thaliana. Intact DREB2A expression does not activate downstream genes under normal growth conditions. A negative regulatory domain exists in the central region of DREB2A, and deletion of this region transforms DREB2A to a constitutive active form (DREB2A CA). We carried out microarray analysis of transgenic… Show more

“…For example, overexpression of DREB2A in transgenic plants could not improve stress tolerance, but overexpression of DREB2ACA, carrying a deletion of the central negative regulatory domain (NRD), activated the expression of many stress-inducible genes and improved tolerance to drought and heat stress in transgenic Arabidopsis (Sakuma et al, 2006a(Sakuma et al, , 2006b. Similarly, AREB1, a homologous gene of OsbZIP46, has been reported to fully function after posttranslational modification (Fujita et al, 2005).…”

“…DREB2A expression is induced rapidly by HS but only gradually induced by drought stress. 8 In eukaryotic cells, the control of gene expression is determined by transcription and by post-transcriptional control. 9 Eukaryotic cells under stressful conditions halt translation initiation and induce formation of cytoplasmic granular structures formed by RNA-protein complexes, which are referred to as stress granules (SGs) or processing bodies (P-bodies).…”

“…It is a member of the DREB subfamily within the APETALA2/ethylene responsive-element binding factor (AP2/ERF) family of transcription factors and contains a single conserved DNA-binding domain (Liu et al, 1998;Sakuma et al, 2002). Its expression is induced by dehydration, high salinity, and heat shock (Liu et al, 1998;Sakuma et al, 2006b). The transcription of DREB2A in response to dehydration and heat shock is independently regulated by different regions in the DREB2A promoter Yoshida et al, 2011).…”
