Dual regulation by ethanol of the inhibitory effects of ketamine on spinal NMDA-induced pressor responses in rats

Abstract: BackgroundAcute exposure of ethanol (alcohol) inhibits NMDA receptor function. Our previous study showed that acute ethanol inhibited the pressor responses induced by NMDA applied intrathecally; however, prolonged ethanol exposure may increase the levels of phosphorylated NMDA receptor subunits leading to changes in ethanol inhibitory potency on NMDA-induced responses. The present study was carried out to examine whether acute ethanol exposure influences the effects of ketamine, a noncompetitive NMDA receptor … Show more

“…To determine the effects of Aβ on cardiovascular functions, we performed the intracerebroventricular injection of Aβ on NMDA receptor function in the RVLM. The resting MAP 81.37 ± 0.81 mmHg (n = 72) in urethane-anesthetized male SD rats was similar to our previous study [36,37]. Consecutive microinjection of NMDA (0.14 nmol, 100 nL) into the RVLM at intervals of 30 min induced an increase in MAP; the pressor responses were onset within 2 min and lasted for 10-15 min.…”

“…The procedure for Western blot analysis of brain tissue was similar to that described in earlier studies [ 32 , 34 ]. Rats were sacrificed 30 min after oral gavage of ethanol or saline.…”

“…Although in that paper we were not able to determine which of the many ion channels regulated by zinc were involved, several channels were raised as candidates. The pharmaceutical formulation of the metal plus vitamin supplements, as currently used, is a correct approach that may result in brain function improvement [ 133 ]. As to the use of nanostructured particles for zinc delivery to improve the pharmacokinetics of its administration, particularly its brain accessibility, it remains as an open question that deserves further experimentation.…”
