Dynamic Assessment of Bilayer Thickness by Varying Phospholipid and Hydraphile Synthetic Channel Chain Lengths

Abstract: A library of "hydraphile" synthetic ion channel analogues that differ in overall length from ∼28-58 Å has been prepared. A new and convenient ion-selective electrode (ISE) method was used to assay Na + release. Liposomes were formed from three different phospholipids: 1,2-dimyristoleoyl-snglycero-3-phosphocholine (DMPC), 1,2-dioleoyl-sn-glycero-3-phosphocholine (DOPC), and 1,2-dierucoyl-sn-glycero-3-phosphocholine (DEPC). The acyl chains of the lipids comprise cisunsaturated 14:1, 18:1, or 22:1 residues, respe… Show more

“…We have used NMR methods 16 and direct detection of Na + release by use of sodium-sensitive ion selective electrodes. 8 We have also used the planar bilayer clamp method to confirm channel-like behavior for these compounds. 17,18 The latter method provides considerable detail about ion transport through the bilayer but it is a complex technique that requires quite specialized instrumentation.…”
