Dynamic Autophosphorylation of Mps1 Kinase Is Required for Faithful Mitotic Progression

Abstract: The spindle assembly checkpoint (SAC) is a surveillance mechanism monitoring cell cycle progression, thus ensuring accurate chromosome segregation. The conserved mitotic kinase Mps1 is a key component of the SAC. The human Mps1 exhibits comprehensive phosphorylation during mitosis. However, the related biological relevance is largely unknown. Here, we demonstrate that 8 autophosphorylation sites within the N-terminus of Mps1, outside of the catalytic domain, are involved in regulating Mps1 kinetochore localiza… Show more

“…Upon inhibition of kinase activity, Mps1 localization on kinetochores is elevated significantly ( Fig. S4 A and B) (22,35,44,45). Therefore, we hypothesized that the abnormal accumulation of inactive Mps1 in kinetochores causes defects in kinetochore-MT attachments.…”

“…Different studies have implicated Mps1 in chromosome alignment, but the underlying molecular mechanism remains unclear [336,337,338,339]. Initially it was proposed that regulation of chromosome alignment by Mps1 acts through modulation of Aurora B kinase activity [337].…”
