Dynamic behaviour of oxidized glutathione in solution investigated by nuclear magnetic resonance

Abstract

In view of the important biological functions of oxidized glutathione (GSSG), we investigated the correlation between dynamic and conformational properties and biological activity. Anisotropic molecular motion characterizes different GSSG molecular districts. This information, obtained by NMR carbon relaxation investigations, suggests that the peptide does not independently assume any stable structure in solution. Analysis of the effects of the addition of a stable spin label to the solution confirmed the absence of conformation of GSSG in D20. Moreover, the paramagnetic effects observed on proton and carbon nuclei of oxidized glutathione suggest that the dipolar term is the main source of paramagnetic relaxation.CLAUDIO ROSSI, ALESSANDRO DONATI SERGIO, ULGIATI et MARIA ROSARIA SANSONI. Un mouvement molCculaire anisotropique caracterise les differentes portions molCculaires du GSSG. Cette information, obtenue par des etudes de relaxation en RMN du carbone, suggitre que, d'une faqon independante, le peptide n'assume aucune structure stable en solution. L'analyse des effets de l'addition d'un marqueur de spin la solution a confirme l'absence de conformation dans le GSSG dans le D,O. De plus, les effets paramagnetiques observes sur le proton et les noyaux de carbone du glutathion oxyde suggitrent que le terme dipolaire est la principale source de relaxation paramagnetique.[Traduit par la redaction]
