Dynamic Contrast-Enhanced MRI Measurements of Passive Permeability through Blood Retinal Barrier in Diabetic Rats

Abstract: DCE-MRI provides a sensitive, noninvasive, and linear assay that accurately measures, without potential artifacts associated with death and enucleation, passive BRB PS in experimental diabetes. DCE-MRI BRB PS measurements are expected to provide a useful surrogate marker of drug treatment efficacy.

“…All patients were well enough to attend as outpatients. The BBB imaging technique was similar to that used previously in the brain, 15 retina, 16 and breast. 17 We tested 2 temporal modifications of the technique in 2 consecutive cohorts, both including patients with lacunar and cortical ischemic strokes.…”

“…It has been established that DCE-MRI measurements of BRB PS are capable of detecting subtle changes in vascular permeability (2). Further, quantifiable changes in BRB permeability have proved useful for the assessment of various therapeutic interventions, including corticosteroid (triamcinolone acetonide) treatment and anti-vascular endothelial growth factor therapy (2,32).…”

“…To avoid these potentially confounding factors, including the near-death artifact, DCE-MRI was applied to rats that were diabetic for 2, 4, 6, or 8 months (2). No evidence of passive BRB leakage in rats was found until after 6 months of diabetes ( Fig.…”
