Dynamic image denoising for voxel-wise quantification with Statistical Parametric Mapping in molecular neuroimaging

Abstract: PurposePET and SPECT voxel kinetics are highly noised. To our knowledge, no study has determined the effect of denoising on the ability to detect differences in binding at the voxel level using Statistical Parametric Mapping (SPM).MethodsIn the present study, groups of subject-images with a 10%- and 20%- difference in binding of [123I]iomazenil (IMZ) were simulated. They were denoised with Factor Analysis (FA). Parametric images of binding potential (BPND) were produced with the simplified reference tissue mod… Show more

“…Since β must be reliably estimated, ROI-level data are the likely current limit for this approach because the level of statistical noise for voxel data may not yield reliable estimation of β . Denoising algorithms 16,17 may prove useful to improve β estimation, particularly for regions with small volumes. In these data, 30 min of dynamic data were acquired in 5 min bins, allowing for 6 data points to estimate β.…”

“…For 5-HT 2A R binding, parametric images of SUR were compared between groups using the SPM12 software (Wellcome Trust Centre for Neuroimaging, UCL, London, UK) and the Small Animal Molecular Imaging Toolbox 28 (SAMIT, Groningen, Netherlands) in Matlab (R2019, Mathworks Inc, USA). An uncorrected p at 0.001 with a cluster size threshold of 100 voxels was employed 29 , 30 . All the statistical tests were two-sided.…”

“…Radioactive decay correction was performed, while correction for attenuation or scatter was not. Following reconstruction, dynamic images from the partial saturation experiment were denoised with factor analysis (FA) using Pixies software (Apteryx, Issyles-Moulineaux, France) as previously described [23,27,35]. FA allows the decomposition of a dynamic signal into a few elementary components, termed factors [27,28,36].…”
