DZNep inhibits the proliferation of colon cancer HCT116 cells by inducing senescence and apoptosis

Abstract: EZH2 is over-expressed in human colon cancer and is closely associated with tumor proliferation, metastasis and poor prognosis. Targeting and inhibiting EZH2 may be an effective therapeutic strategy for colon cancer. 3-Deazaneplanocin A (DZNep), as an EZH2 inhibitor, can suppress cancer cell growth. However, the anti-cancer role of DZNep in colon cancer cells has been rarely studied. In this study, we demonstrate that DZNep can inhibit the growth and survival of colon cancer HCT116 cells by inducing cellular s… Show more

“…Targeting EZH2 can be an effective therapeutic strategy for colon cancer. Sha et al demonstrated that DZNep can inhibit the growth and survival of colon cancer HCT116 cells by inducing cellular senescence and apoptosis (9). This study is also consistent with our results in which DZNep showed a toxic activity on HT-29 cell line.…”

“…3-Deazaneplanocin A (DZNep) is an S-adenosylhomocysteine synthesis and histone methyltransferase EZH2 inhibitor that has a wide range of effects in various human cancers. In recent studies, DZNep has also been shown to have an impact on apoptosis in CRC cell cultures (9). However, the relationship of AP-related genes in DZNep-treated CRC cells remains to be elucidated.…”

“…Abnormal proliferation is a common feature of malignant tumors. DZNep has been confirmed to be able to inhibit the proliferation of Eca109 oesophageal squamous cell carcinoma cells and HTC116 colorectal cancer cells (25,26). The present study used the poorly differentiated human BGC-823 gastric adenocarcinoma cell line.…”

“…16, 34 Our previous work also proved the senescence-inducing ability of an EZH2 inhibitor DZNep in HCT116 cells. 35 Although p16 induction is not observed, p21 is indeed regulated by EZH2 and contributes substantially to senescence induction after DZNep or LDM treatment. Meanwhile, the p21 induction by LDM occurs independently of p53.…”
