EAE Tolerance Induction with Hsp70-Peptide Complexes Depends on H60 and NKG2D Activity

Abstract: Inflammation leads to induction of tissue stress conditions that might contribute to the generation of mechanisms limiting ongoing immune responses. We have shown previously that peptides derived from brain tissue of mice with experimental autoimmune encephalomyelitis (EAE) complexed with the chaperone heat shock protein 70 (Hsp70-pc) induce an NK-cell-dependent tolerance for subsequent EAE sensitization. We now present data that showed that the MHC class I-related glycoprotein H60 determines Hsp70-pc-induced … Show more

“…Thus, NKG2D-ligand system works in a broad range of diversity but mounts an efficient immune response. Recently the instrumental role of the Hsp70 ⅐ peptide complex in NKG2D-mediated IFN-␥ secretion and its active role in the induction of tolerance against experimental autoimmune encephalomyelitis was demonstrated (41). Our observations about a 110-kDa Hsp working as a NKR-P2 ligand on rat DCs provides the evidence of additional diversity for an efficient tumor recognition system that is also supported by the binding of the Hsp70 ⅐ peptide complex to NKG2D.…”

“…Induction of H60 as an NKG2D ligand has been reported with several different models including transplantation (40, 41). Interestingly, BALB.B donor T cells upregulated surface H60 expression after activation both in vitro and in vivo.…”

“…H60a can mediate strong suppressive effects on T cell proliferation [30]. It has also been shown to play a role in NK cell tolerance in experimental autoimmune encephalomyelitis [31]. Additionally, a recent study, using a mouse model of spontaneous autoimmune arthritis [32], provided evidence suggesting that efficient suppression of autoimmune diseases may require polyclonal specificities rather than single antigen-specific regulatory T cell activity.…”
