EAF2 Suppresses Hypoxia-Induced Factor 1α Transcriptional Activity by Disrupting Its Interaction with Coactivator CBP/p300

Abstract: bPrevious studies revealed that the potential tumor suppressor EAF2 binds to and stabilizes pVHL, suggesting that EAF2 may function by disturbing the hypoxia signaling pathway. However, the extent to which EAF2 affects hypoxia and the mechanisms underlying this activity remain largely unknown. In this study, we found that EAF2 is a hypoxia response gene harboring the hypoxia response element (HRE) in its promoter. By taking advantage of the pVHL-null cell lines RCC4 and 786-O, we demonstrated that hypoxia-indu… Show more

“…SHARP1 promotes HIF-α degradation by serving as the HIF-presenting factor to the proteasome [26]. EAF2 is a specific HIF-1α downstream target, which interacts with HIF-1α but not HIF-2α, resulting in the inhibition of HIF-1α transcriptional activity via preventing p300 from binding to HIF-1α [27]. Other proteins do not interact with HIF-α directly, but interact with other factors that regulate HIF-α, e.g., pVHL and PHDs among others.…”

“…The immunoprecipitate and the total cell lysate were boiled with 13 SDS sample loading buffer, separated on SDS-PAGE, and transferred to a polyvinylidene difluoride membrane (Millipore). Western blot analysis was performed as described previously (39). Anti-Myc Ab-conjugated agarose beads were purchased from Sigma-Aldrich.…”
