EAPP modulates the activity of p21 and Chk2

Abstract: Genomic instability is thought to be critical for the development of cancer. Among its causes microsatellite instability (MIN) and chromosomal instability (CIN) have attracted the most attention. Cell cycle checkpoints and DNA repair mechanisms are the first line of defense against DNA damage. Among the most dangerous DNA lesions are double-strand breaks. The response to DNA double strand breaks is regulated mainly by the serine/threonine kinases ATM and Chk2 and their downstream target the tumor suppressor p5… Show more

“…In addition, USF1 levels did not differ between Trp53 KD cells and controls, indicating that USF1 expression is not dependent on p53 ( Figure 2 , A and B). This also suggests that the deficiency in cell cycle arrest of Usf1 KD cells in response to genotoxic stress may be the result of the absence of increased levels and/or activity of p53 and p21 [5] , [6] , [7] , [30] , [31] .…”

“…4B, p21 expression levels were increased when cells contained the BGLF2 expression plasmid relative to the empty plasmid control, confirming that BGLF2 can induce p21 independently from p53. The expression of E2F-associated phosphoprotein (EAPP) is also important for activating p21 transcription and can act independently of p53 (40,41). Therefore, we also examined possible effects of BGLF2 on EAPP levels; however, BGLF2 did not significantly alter EAPP levels relative to those of the empty plasmid control (Fig.…”

“…The cyclin-dependent kinase inhibitor p21 is a target of p53 and can be stimulated under oncogenic stress. p21 upregulation was also demonstrated under DNA damage conditions due to an aberrant ratio of Sp1 and E2F transcription factors [11]. Thus, EAPP may be involved in the regulation of self-protective cellular functions in the DNA damage response and anti-apoptotic roles by stimulating p21 protein expression.…”
