Early bronchiolar damage following paraquat poisoning in mice

Abstract: Paraquat causes focal intracellular oedema of the terminal bronchiolar epithelial cells and focal subpleural atelectasis with thickening of the interalveolar septa 1 hr after the administration of an LD50. These changes are progressive, and lead to panacinar atelectasis with necrosis plus sloughing of epithelial cells in many terminal bronchioles. Radioactive phosphatidyl choline (PC) is recoverable by lavage within 90 s of the administration of tritiated palmitate, which supports previous suggestions that one… Show more

“…Acrolein, a degradation product ofcyclophosphamide (16), is also moderately toxic (TD50 = 100 MM) in the in vitro system. Another drug, busulfan, has little effect on Clara cell attachment efficiency at doses < 100 AM, a finding that is similar to that with alloxan, an agent considered to selectively damage lung endothelial cells (18).…”

“…It is one of the most toxic poisons involved in suicide attempts. Paraquat primarily causes lung injury in mammals, where paraquat is accumulated through a process of active transport in Clara cells and alveolar type I and II epithelial cells [3][4][5][6][7]. There are two distinct phases in the development of pulmonary fibrosis by paraquat: an early destructive phase characterized by alveolitis, pulmonary edema, and infiltration of inflammatory cells, followed by a final fibrotic phase where fibroblasts proliferate, collagen is deposited, and pulmonary fibrosis occurs [3,[8][9][10].…”
