Early decrease of serum biomarkers of type II collagen degradation (Coll2‐1) and joint inflammation (Coll2‐1 NO2) by hyaluronic acid intra‐articular injections in patients with knee osteoarthritis: A research study part of the Biovisco study

Abstract: To measure the evolution of the serum levels of specific Osteoarthritis (OA) biomarker, Coll2-1 and Coll2-1 NO 2 in knee osteoarthritic patients after viscosupplementation (VS). Fifty-one patients with unilateral symptomatic knee were recruited for this prospective open label study. They received three intra-articular injections of 2 ml of hyaluronic acid (Hylan GF-20) and were followed for 3 months. Walking pain was evaluated and serum samples were taken at each visit. Coll2-1 and Coll2-1 NO 2 were measured i… Show more

“…HA injections in a previous study including 45 patients produced a decrease in Coll2-1 superior to 10 nmol/l in 48% of the patient [20]. The hypothesis was made that Kartilage® Cross treatment in this study will produce a decrease of 10 nmol/l of Coll2-1 between inclusion and D90 in the serum in 45% of the patients.…”

“…When measured in serum of patients with OA it was found to be in higher concentrations than age-matched controls [18]. Interestingly, Coll2-1 has been shown to be decrease in serum of patients with knee OA after a series of 3 injections of hylan GF-20, suggesting that this biomarker could be helpful for the monitoring of IAHA effect [20]. Coll2-1 is a biomarker of OA that entered the qualification process [21].…”

“…Proteoglycan and type II collagen are degraded at the surface, leading to loss of structural strength of articular cartilage due to aggrecanase and matrix metalloproteinases (MMPs) (Struglics et al 2006) and decreased type II collagen and aggrecan integrity (Garnero et al 2002;Jalbă et al 2011;Henrotin et al 2013). Therefore, type II collagen and proteoglycan can be used as biochemical markers of early stage canine OA (Matyas et al 2004).…”

“…The more probable hypothesis is a transient restoration of the physiological joint homeostasis [6]. Moreover there are increasing in vitro and in vivo evidences that HA could have chondroprotective properties [7][8][9][10][11][12] whereas metaanalyses continue to show controversial results regarding the efficacy and safety of viscosupplementation [13][14][15][16][17].…”
