Early onset adult deafness in the Rhodesian Ridgeback dog is associated with an in-frame deletion in the EPS8L2 gene

Abstract: Domestic dogs exhibit diverse types of both congenital and non-congenital hearing losses. Rhodesian Ridgebacks can suffer from a progressive hearing loss in the early stage of their life, a condition known as early onset adult deafness (EOAD), where they lose their hearing ability within 1–2 years after birth. In order to investigate the genetic basis of this hereditary hearing disorder, we performed a genome-wide association study (GWAS) by using a sample of 23 affected and 162 control Rhodesian Ridgebacks. W… Show more

“…At this point, we wanted a simple mechanism by which to positively identify our cells as being related to the original tumors from which they were derived, and we turned to genotype analysis. The CanineHD beadchip (Illumina Inc.) contains approximately 220,000 single nucleotide polymorphisms (SNPs) across the canine genome 55 . We collected DNA from tumor samples, PDX-samples, and cell line samples, and submitted the DNA for genotyping.…”

“…Identification of component ancestry for mixed breed dogs is also big business. The widespread use of these tests is helping researchers to characterize the broad population-based allele frequencies associated with many diseases and to also help map new disease risk variants ( Donner et al 2018 , 2023 ; Kawakami et al 2022 ).…”

“…A particular type of early onset adult deafness was identified in Rhodesian Ridgebacks, with occurrence within the first 1-2 years of life. The associated variant was a 12 bp deletion in the EPS8L2 gene [143]. In Rottweilers, sensorineural bilateral deafness with onset at a few weeks of age was associated with a missense variant in the LOXHD1 gene [144].…”
