Early Reactions of the Subcutaneous Tissue to Repeated Injections of Carcinogens in Aqueous Solutions

Abstract: SUMMARY.-Four water-soluble carcinogens were injected at the same site subcutaneously into rats, mice or guinea-pigs, twice weekly for 5-8 weeks in order to study the evolution of the early tissue reaction. MNU was injected into rats as 0-1 ml. of 0.5% solution, and into mice as 0-1 ml. of 0-05% solution. NQO was administered to rats (0-1 or 0-2 ml. of 0.25 or 0-1 %) mice (0-1 of 0-05%) and guinea-pigs (0-5 of 0.1%). Propane sultone and BEI were administered to rats only, the former as 0-1 ml. of 3% and the la… Show more

“…This particular aspect of carcinogen treatment has also been noted by Vasiliev et al (1962), who demonstrated an inhibition of fibroblastic differentiation and growth around paraffin pellets containing carcinogens. Similar findings (Hooson et al, 1971(Hooson et al, , 1973 followed repeated injections of watersoluble carcinogens into the subcutis. Moreover, carcinogenic hydrocarbons inhibit the proliferation of fibroblasts in vitro, whilst chemically related but noncarcinogenic hydrocarbons do not (Vasiliev & Guelstein, 1963).…”

“…This led to a profound distortion of normal fibrogenesis, and encapsulation of the implanted material. Similar responses were seen by Hooson et al (1971Hooson et al ( , 1973 after s.c. injection of water-soluble carcinogens.…”

“…Leaving frank carcinogens aside, it was noted that the otherwise biologically inert materials which did lead to intense tissue reactions and injection site sarcomas were those which possessed surface activity, acidic pH or precipitated at the site of injection [103, 107–110]. …”

“…Histological descriptions of the connective tissue reaction around silicone rubber implants (Nothdurft, 1961) and various plastic films (Oppenheimer et al, 1959) emphasize the importance of the formation of a thick, avascular connective tissue capsule, with consequent isolation of the fibroblasts, as an essential step in the evolution of malignancy.…”
