Neural necrosis virus (NNV) of family Nodaviridae affect wide range of fish species with viral encephalopathy and retinopathy causing mass mortality up to 100%. Currently there is no effective treatment and depopulation is only suggested recommendation. New avenues and approach are required to control this harmful malady. In this study we developed an epitope-based vaccine (EBV), against NNV using computation approach. We have selected two conserved proteins RNA-dependent RNA polymerase (RdRP) and capsid proteins. Based on more than ~ 1000 epitopes we selected six antigenic epitopes. These were conjugated to adjuvant and linker peptides to generate a full-length vaccine candidate. Biochemical structural properties were analyzed by Phyre2 server. ProtParam, Molprobity. Ramachandran plot results indicate that 98.7% residues are in a favorable region and 93.4% residues in the favored region. The engineered EBV binds to toll like receptor-5 (TLR5) an important elicitor of immune response. Further molecular docking by PatchDock server reveals the atomic contact energy (i.e. − 267.08) for the best docked model of EBV and TLR5 receptor. The molecular simulation results suggest a stable interaction; the RMSD and RMSF values are 1–4 Ǻ and 1–12Ǻ, respectively. Further we have suggested the best possible codon optimized sequence for its cloning and subsequent purification of the protein. Overall, this is a first report to suggest an in-silico method for generation of an EBV candidate against NNV. We surmise that the method and approach suggested could be used as a promising cure for NNVs.

Mucosal immunity plays a critical role in the protection of teleost fish against infection, but mucosal immunoglobulin of important aquaculture species unique to Southeast Asia remained greatly understudied. In this study, the sequence of immunoglobulin T (IgT) from Asian sea bass (ASB) is described for the first time. IgT of ASB possesses the characteristic structure of immunoglobulin with a variable heavy chain and four CH4 domains. The CH2-CH4 domains and full-length IgT were expressed and CH2-CH4 specific antibody was validated against full-length IgT expressed in Sf9 III cells. Subsequent use of the anti-CH2-CH4 antibody in immunofluorescence staining confirmed the presence of IgT-positive cells in the ASB gill and intestine. The constitutive expression of ASB IgT was characterized in different tissues and in response to red-spotted grouper nervous necrosis virus (RGNNV) infection. The highest basal expression of secretory IgT (sIgT) was observed in the mucosal and lymphoid tissues such as the gills, intestine and head kidney. Following NNV infection, IgT expression was upregulated in the head kidney and mucosal tissues. Moreover, a significant increase in localized IgT was found in gills and intestines of infected fish on day 14 post-infection. Interestingly, a significant increase in NNV-specific IgT secretion was only observed in the gills of the infected group. Our results suggest that ASB IgT may play an important role in the adaptive mucosal immune responses against viral infection and could potentially be adapted as a tool for the evaluation of prospective mucosal vaccines and adjuvants for the species.

Indonesia's fish production is abundant, especially in aquaculture. Lates calcarifer is farmed fish species. The failure of L. calcarifer cultivation due to disease problems. We are utilizing a natural compound derived from tropical Sargassum sp. extracts, namely alginate. This study aims to determine and analyse the supplementation of alginate in the diet by oral administration to improve the fish's hematological performance and oxygen stress tolerance. There were one control and three treatments (2 g.kg-1, 4 g.kg-1, and 8 g.kg-1 alginate/feed). The experiment was a completely randomized design with three replications. Fish were reared in a 350 L fiber tank for 12 days at a density of 20 ind.tank-1. Stress oxygen tolerance was applied by rearing the 10 fish in 12 L fully plastic-wrapped containers. The parameters test analyzed were phagocytic activity and phagocytic index, red blood parameters which are hematocrit and hemoglobin. The survival rate after 5 hr anoxic exposure was also recorded. The best treatments were achieved at a dose of 6 g.kg-1 and 8 g.kg-1in all parameters, except the phagocytic index. The alginate addition at all oxygen stress treatments also performed a better survival rate compared to the control. Adding alginate to feed as feed supplementation by oral administration can boost hematological performance and higher tolerance from oxygen stress exposure.
