EcoTILLING in Beta vulgaris reveals polymorphisms in the FLC-like gene BvFL1that are associated with annuality and winter hardiness

Abstract: BackgroundSugar beet (Beta vulgaris ssp. vulgaris L.) is an important crop for sugar and biomass production in temperate climate regions. Currently sugar beets are sown in spring and harvested in autumn. Autumn-sown sugar beets that are grown for a full year have been regarded as a cropping system to increase the productivity of sugar beet cultivation. However, for the development of these “winter beets” sufficient winter hardiness and a system for bolting control is needed. Both require a thorough understandi… Show more

“…EcoTILLING is shown to be a fast, convenient, and effective method for detecting SNPs in genes of haploid species, such as fungus. The method has been widely adapted for many crop species [43][44][45]. Having established EcoTILLING for C. sativus, we expect that it can be similarly adapted for many microorganisms.…”

“…The sequence variations (Substitutions and Indels) were detected by using ‘Multalin’ software (http://multalin.toulouse.inra.fr/multalin/, Corpet (1988). The SNP frequency was calculated using the formula: (Total number of SNPs detected / total length of the screened fragment) X 1000 (Frerichmann et al ., 2013). PARSESNP (Taylor and Greene, 2003) was used to analyze and display the variation in gene sequences, and SIFT (Sorting intolerant from tolerant; Sim et al ., 2012) was used to predict the deleterious variations for protein functions.…”

“…This technique is now applied to rice, maize, lotus, poplar , Brassica, zebrafish, Drosophila, Caenorhabditis and human , indicating its broad applicability. The implication of ECO-TILLING to identify potential novel functional alleles in the known and candidate genes/transcription factors (TFs) regulating qualitative and quantitative agronomic traits by association/genetic mapping is well-documented for expediting the genetic enhancement of crop plants (Mejlhede et al, 2006;Barkley and Wang, 2008;Ibiza et al, 2010;Negrao et al, 2011;Yu et al, 2012;Frerichmann et al, 2013). ECO-TILLING usually employs a mismatchspecific CEL-I nuclease to cleave the PCR amplified fragments at the site of heteroduplex formation involving nucleotide (SNP-allelic) polymorphism.…”
