Effect of 5‐Hydroxytryptamine on Steroidogenesis and Oocyte Maturation in Pre‐ovulatory Follicles of the Medaka Oryzias latipes

Abstract: The effect of 5-hydroxytryptamine (5-HT) on steroidogenesis and oocyte maturation in pre-ovuiatory follicles of the medaka Oryzias lafipes was examined using in vitro culture system. The earliest breakdown of the germinal vesicle of intrafollicular oocytes occurred about 17 hr after the beginning of incubation in the presence of 5-HT at concentration of 10 ng/ml or more. 5-HT induced oocyte maturation in a dosedependent manner. Cyanoketone inhibited this stimulation. The concentration of 5-HT required to induc… Show more

“…In contrast to the observed inhibitory influence in killifish, 10 µg/L and 100 µg/L of 5-HT had a stimulatory effect on oocyte maturation in Japanese medaka in vitro by inducing synthesis of estradiol and MIS in granulosa cells, resulting in germinal vesicle breakdown. 164 Overall, these studies demonstrate the complexity of serotonergic actions involved in the control of the HPG axis in fish. Moreover, the modulatory function of 5-HT appears to be largely dependent on reproductive status and may vary considerably in different fish species.…”

“…The SSRIs also have been measured in several species of fish collected from municipal effluent-dominated streams [2,11], indicating the potential problem that SSRIs may impose on the environment. Serotonin is assumed to be an important, ubiquitous neuromodulator in all vertebrates and invertebrates, and studies indicate that increased 5-HT concentrations stimulate gonadotropin secretion from the pituitary gland of the Japanese medaka, Oryzias latipes, which further increases gonad steroidogenesis [4,12]. In another fish species, Fundulus heteroclitus, increased levels of 5-HT inhibited oocyte maturity [13], but the response of oocytes to 5-HT varies among species [12][13][14].…”

“…Ultrastructural re-organization involves the redistribution of microfilaments and microtubules that in turn support the relocation of cytoplasmic organelles such as cortical granules and mitochondria [Schorderet-Slatkine 1972;Masui 1985;Iwamatsu et al 1993;Voronina and Wessel 2003;Sun and Schatten 2006;Stricker 2006;Ferreira et al 2009;Liu et al 2010; De los Reyes et al 2011].…”
