“…Thirty min later, when urinary flow rate had decreased markedly, 20 &kg body wt DDAVP (Ferring Sveden) was infused iv over a period of 20 min. Even at such supramaximal doses, the drug has no significant vasoconstrictor effects (26). Clearance measurements as described above, were then performed for at least 3 hr.…”

“…It is well known that an analogue of authentic arginine vasopressin (AVP), 1-deamino-[8-D-arginine] vasopressin(dDAVP), has a greater antidiuretic potency and a much less vasopressor activity than AVP (Huguennin et al, 1965;Vavra et al, 1968;Sawyer et al, 1974-a, b). Although the enhanced potency of dDAVP has been shown to be due to its longer plasma half-life (Edwards et al, 1973;Sawyer et al, 1974-a) and greater generation of renomedullary adenylate cyclase and cyclic AMP than those of AVP (Seif et al, 1978), mechanisms responsible for the prolonged plasma half-life of dDAVP have not yet been fully clarified.…”

“…An analogy may be found in ADH, where replacement of L-arginine by D-arginine in position 8 reduces pressor but not antidiuretic potency (10), and in ACTH where removal of C-terminal 14-39 abolishes corticotropic but enhances melanotropic activity (11) because of possible idiosyncratic or allergic reaction to intravenous injection of the chemically altered forms of native hGH, this type of experiment was not done. It is tempting to speculate that the increased retention of phosphorus seen in man with RCAMhGH and its plasmin digest is related to the enhanced activity of plasmin digests of native hGH (PD-hGH) when assayed in the rat by the tibia test (17)(18)(19).…”
