Effect of Aerobic Exercise on miRNA-TLR4 Signaling in Atherosclerosis

Abstract: Toll-like receptor 4 (TLR4)-tumor necrosis factor receptor 6 (TRAF6) signaling is activated in atherosclerosis (AS), inducing inflammatory mediators. Because miR-146a, a TLR4 microRNA (miRNA), can regulate TLR4 signaling during inflammatory responses, this study investigated the effects of aerobic exercise on TLR4-targeted miRNAs in AS. Apolipoprotein E-null mice fed a high-fat diet for 12 weeks were separated into 3 groups: (i) no treatment (AS), (ii) statin treatment (AD), or (iii) aerobic exercise (AE). Pla… Show more

“…However, they detected normalization of reduced skeletal muscle miRNA-126 levels in spontaneously hypertensive rats (SHR) in response to the protocol, which was paralleled by revascularization and a reduction of blood pressure (Fernandes et al, 2012). In male ApoE null C57BL/6 J mice, miR-126 levels were reported to be increased in vascular tissue after a 12-week treadmill protocol (Wu et al, 2014). In humans, Uhlemann et al (2014) reported plasma concentrations of miR-126 after different single exercise bouts.…”

“…By activating the HAECs with HG/thrombin, we observed increased expression of these CAMs, while the miR-146a mimic attenuated expression of these CAMs and associated THP-1 adhesiveness. Interestingly, Wu et al reported that aerobic exercise and statins could increase miR-146a levels in ApoE-null mice, thereby attenuating vascular inflammation through reducing TLR4 and TRAF6 signaling [52]. Our previous work also found that angiotensin-(1–7) can decrease endothelial IL-6 expression in glycated albumin-stimulated HAECs through its ability to enhance endogenous miR-146a levels [15].…”

“…Increased miR-146a and miR-126 and reduced miR-155 were induced by 12-week aerobic exercise on the aorta of mice with atherosclerosis [182]. Healthy rats subjected to 10-week swimming training showed increased cardiac capillary fiber ratio along with elevation of miR-126 expression, and this promoting angiogenesis effect might be regulated by MAPK and PI3K/Akt/eNOS signaling through targeting Spred-1 and PI3KR2 by miR-126 [183], which has been reported as an endothelial-specific modulator on vascular integrity, angiogenesis and repair [s19][62,64].…”
