Effect of ALDH2 and CYP2E1 Gene Polymorphisms on Drinking Behavior and Alcoholic Liver Disease in Japanese Male Workers

Abstract: The ALDH2 genotype is strongly associated with individual alcohol drinking behavior and the development of alcoholic liver disease in Japanese male workers, but the CYP2E1 genotype is not.

“…Chang et al [23] found that as the key enzyme of alcohol metabolism, ALDH2 gene could influence the individual alcohol consumption and drink frequency. Okamoto et al [24] found that in the Japanese men, the heterozygous GA genotype of ALDH2 gene carriers had the less capacity for alcohol than the wild homozygous GG genotype, a result that Yu et al [25] replicated in the Chinese Han population. The accumulation of acetaldehyde secondary to the reduced ALDH2 enzymatic activity can cause flushing, tachycardia and nausea more rapidly in an individual with the gene polymorphism and effectively restrict alcohol consumption.…”

“…Interestingly, another polymorphism in CYP2E1 , Taq I, is associated with reduced susceptibility to ALD, although this allele does not directly affect alcohol metabolism 69 . Several studies from Japan and Korea have searched for associations between the polymorphisms in CYP2E1 and ALD without positive results 70, 71 . A study of Chinese of Han, Mongol, and Chaoxian nationalities found a positive association between the c2 genotype and ALD 72 .…”

A single nucleotide polymorphism (SNP) located in the promoter region of the TNF-␣ gene, which results in increased cytokine production, is associated with more severe steatohepatitis.…”
