Effect of Amentoflavone on the Inhibition of Pulmonary Metastasis Induced by B16F-10 Melanoma Cells in C57BL/6 Mice

Abstract: This study was an investigation of the antimetastatic activity of amentoflavone using B16F-10 melanoma-induced experimental lung metastasis in C57BL/6 mice. Amentoflavone treatment significantly reduced tumor nodule formation accompanied by reduced lung collagen hydroxyproline, hexosamine, and uronic acid levels. Serum sialic acid and γ γ-glutamyl transpeptidase levels were also significantly inhibited after amentoflavone treatment. Amentoflavone treatment up-regulated the lung tissue inhibitor of metalloprote… Show more

“…These biflavones are rare flavones, comprised of two identical or nonidentical flavonoid units conjoined in a symmetrical or unsymmetrical manner through an alkyl or an alkoxy-based links of varying length (Aravind et al 2008), including 3 0 , 8 00 -linked (Zheng et al 2011), 3 0 , 6 00 -linked , 4 0 -O-8 00 -linked (Dora and Edwards 1991), 4 0 -O-6 00 -linked (Chen, Duh, and Chen 2005;Swamy et al 2006;Zhang et al 2006), and 3-O-4 000 -linked (Zhang et al 2006), 3 0 , 3 000 -linked (Swamy et al 2006), 3 0 , 4 000 -linked series (Swamy et al 2006), and others (Lim et al 2009). They show strong bioactivities, such as vasorelaxation, antifungal, antiviral, antiinflammatory, antioxidant, anticancer bioactivities and inhibition of tumor metastasis (Ma et al 2001;Kang et al 2004;Chen et al 2005;Jung et al 2006;Guruvayoorappan and Kuttan 2007;C. “…Propidium iodide (PI) and RNase were purchased from Biovision (Mountain view, CA, USA) and Thermo Fisher Scientific, respectively. U-87 MG cells (5×10 5 ) were seeded in 6-well plates and treated with 50 or 100 μM amentoflavone for 48 h before harvesting. Cells were then fixed with 70% ethanol overnight and stained with PI/RNase solution (40 μg/ml PI, 100 μg/ml RNase and 1% Triton X-100 in phosphate-buffered saline (PBS) for 30 min.…”

“…Amentoflavone, a polyphenolic compound isolated from Selaginella tamariscina, has been demonstrated to possess anticancer effects through the inhibition of molecules that are associated with tumor progression and modulation of apoptosis (11)(12)(13). Amentoflavone, as a NF-κB signal inhibitor, induces anti-angiogenic and anti-metastatic effects via suppression of NF-κB activation in breast cancer and melanoma cells in vitro and in vivo (11,12).…”
