Pennsylvania (PA) State Racing Commissions regulate the endogenous androgenic steroid, testosterone (TES), in racing intact males (RIM) by quantification of TES in post-race samples. Post-race plasma samples (2209) collected between March 2008 and November 2010 were analyzed for TES, nandrolone (NAN), and other anabolic steroids (ABS). Of the 2209 plasma samples, 2098 had quantifiable TES ≥ 25 pg/mL. Plasma (mean ± SD) concentrations of TES and NAN in RIM were 329.2 ± 266.4 and 96.0 ± 67.8 pg/mL, respectively. Only 64.6% of RIM had quantifiable concentration of NAN, and there was no relationship between TES and NAN. Plasma TES concentrations were significantly (P < 0.0001) higher during the months of April, May, June, July, and August. A significantly higher (P < 0.006) plasma TES was observed in Thoroughbred (TB) (347.6 ± 288.5 pg/mL) vs. that in Standardbred (STB) (315.4 ± 247.7 pg/mL). Plasma concentrations of TES from breeding stallions (BS) were 601.6 ± 356.5 pg/mL. Statistically significant (P < 0.0001) lower plasma concentrations of the two steroids were observed in RIM horses. Based on quantile distribution of TES in the RIM and BS populations, 99.5% were at or below 1546.1 and 1778.0 pg/mL, respectively. Based on this population of RIM, the suggested upper threshold plasma concentration of endogenous TES in horses competing in PA should remain at 2000 pg/mL.

Different anabolic steroids can exercise different effects on the pituitary-gonadal axis in males. During a pilot study regarding the possible beneficial effect of the anabolic steroid nandrolondecanoate (ND) on bone metabolism in patients with rheumatoid arthritis additional endocrinological parameters were studied. A significant decrease was found in the serum levels of testosterone, androstenedione and FSH and the ratio of testosterone/oestradiol. There was a significant increase in the serum levels of oestrone. The levels of oestradiol, SHBG, LH and cortisol remained unchanged. An inhibitory effect of ND on testicular testosterone secretion is assumed. The decrease in androstenedione levels is explained by the diminished testosterone secretion. The rise in oestrone levels is explained by peripheral aromatizing of ND to oestrogens. The presented findings are in accordance with the hypothesis that sex steroids can act directly on the pituitary resulting in selective FSH and LH secretion. The possible role of the ratio testosterone/oestradiol in controlling gonadotrophin output is discussed.Anabolic steroids have been used, and probably are still being used, in the treatment of rheumatoid arthritis. It is thought that, apart from other ef¬ fects, e.g. a general euphoric effect, anabolic ste¬ roids have a beneficial effect on the osteoporosis associated with rheumatoid arthritis. The sugges¬ tion that anabolic steroids have a positive effect on bone metabolism is based on the established role of oestrogen administration in post-menopausal osteoporosis (Lindsay et al. 1976). A similar role is assumed for androgen administration in male osteoporosis. In androgen deficiency syndromes complicated by osteoporosis androgen therapy has been shown to be effective (Baran et al. 1978), but a role for androgens in osteoporosis unrelated to male hypogonadism is not established (Chesnut et al. 1977). In a pilot study regarding the possible effect of anabolic steroids on bone metabolism in patients with rheumatoid arthritis (Bijlsma et al.1982) the testosterone derivative nandrolondeca¬ noate (ND) was used. ND is a 19 norsteroid (17ß-hydroxy-4-oestron-3-one,17-decanoate), which has a widespread use, e.g. in the treatment of anaemia in patients receiving maintenance haemodialysis (Neff et al. 1981). The pharmacological properties of ND are described by de Visser & Overbeek (1960). More recently it became possible to meas¬ ure the levels of sex hormones, gonadotrophins and sex hormone binding globulin by radioimmu¬ noassays. Studies have been done with other anabolic steroids. The data concerning a decrease in testo¬ sterone levels correspond for all steroids used. However, the influence of the different anabolic steroids on the gonadotrophins differs. Aakvaag & Str0mme (1974) found no effect on LH and FSH using mesterolone. Kilshaw et al. (1975) found a decrease in LH and FSH in one patient using methandrostenolone. The effect of a low-dose treatment (10 mg/day) of stanozolol on the anaerobic threshold (AT) and on maximal velocity (Vmax) was studied in ten well-trained runners in a 6-week trial experiment. The serum levels of testosterone (T), interstitial cell-stimulating hormone (ICSH), and of follicle-stimulating hormone (FSH) were determined before, during, and after the steroid administration. No improvements of AT and Vmax were found. No improvement of the competition running times was observed during the experimental period. The ICSH and FSH serum concentrations did not change significantly, while a marked decrease of the T serum concentration was observed. It is concluded that a low-dose treatment of stanozolol, sufficient to depress the testicular function, does not improve athletic performance.
