The aim of the present study was to evaluate the effect of the addition of two olive oil‐derived antioxidants, hydroxytyrosol (3,4‐dihydroxyphenylethanol, HT) and 3,4‐dihydroxyphenylglycol (DHPG), on ovine semen during liquid storage at 5°C and 15°C. Semen was collected, pooled, diluted and then divided into aliquots supplemented with different concentrations (5 μg/ml, 10 μg/ml, 50 μg/ml and 100 μg/ml) of HT, DHPG and a mixture (MIX) of both antioxidants. Sperm motility characteristics were assessed in the different samples at 0, 6, 24, 48, 72 and 96 hr after cooling, and a fertility trial was also conducted. The results showed that the antioxidant addition did not significantly improve total and progressive motility in ovine cooled sperm maintained at 15° or 5°C. However, in samples stored at 5°C, LIN (48, 72, 96 hr), STR (0 hr) and WOB (0, 48, 72, 96 hr) values significantly decreased in comparison with control treatment when high antioxidant concentrations were added (MIX100 or HT100). When samples were maintained at 15°C, MIX50 showed significantly higher VCL values than the control treatment after 6 hr cooling, and MIX100 showed significantly lower VCL values at 96 hr after cooling. According to the artificial insemination trial, no significant differences were observed when antioxidants were added. In conclusion, the use of HT and DHPG showed small impact in sperm motility and fertility was not affected (nor detrimentally nor positively) when insemination was carried out using antioxidant‐supplemented liquid sperm.

Adding a certain amount of antioxidants to semen extender has been shown to improve semen quality. The aim of present study was to elucidate whether the supplementation of melatonin to the Tris-based extender (CTR) could enhance the quality of ram spermatozoa during storage at 4°C. Ram semen samples were collected and diluted with CTR extender containing different concentrations (0, 0.05 (M 0.05), 0.1 (M 0.1), 0.2 (M 0.2) or 0.4 (M 0.4) mM) of melatonin. Sperm routine indicators, mitochondrial activity, total antioxidant capacity (T-AOC) and malondialdehyde (MDA) content were analysed in control and melatonin treatment groups. The higher per cent of motility, plasma membrane integrity, mitochondrial activity and T-AOC activity was observed in M 0.05, M 0.1 and M 0.2 groups compared to control group at 5 days of storage (p < 0.05), while lower percentage of MDA content was observed among these groups (p < 0.05). In addition, there were no significant differences in acrosome integrity among the control and M 0.05, M 0.1 and M 0.2 groups during the experiment. The above results show that the addition of 0.05, 0.1, 0.2 mM of melatonin is beneficial to the preservation of ram semen during liquid storage at 4°C mainly through antioxidative stress. This study aimed to evaluate the effect of addition of aqueous or methanolic Moringa oleifera leaves extract (MOLE) at different levels (0, 100, 200 and 300 µg/ml) as antibiotic alternative to Friesian bull semen extender on sperm characteristics post-dilution, equilibration and thawed. The enzyme activity and antioxidant capacity of bull seminal plasma were determined in post-thawed semen. Five Friesian bulls (3-4 years old and 400-450 kg LBW) were used. Semen was collected from bulls by an artificial vagina twice a week for 5 weeks. After ejaculation semen was evaluated for mass motility and only the ejaculates of ≥70% percentage were pooled and diluted at 37 0 C using citrate-egg yolk extender and divided into seven portions, control (E1), 100 (E2), 200 (E3), 300 (E4) µg aqueous MOLE /ml semen extender or 100 (E5), 200 (E6), 300 (E7) µg methanolic MOLE /ml semen extender. After semen dilution it was equilibrated at 5 0 C for 4 h then it placed in liquid nitrogen at-196 0 C. Frozen semen stored for two month was thawed at 37 0 C for 30 sec. Semen samples were evaluated for characteristics including sperm motility, livability, abnormality, acrosome damage and hypo osmotic swelling test (sperm cells with curled tail) in diluted, equilibrated and thawed semen. Activity of AST, ALT, LDH, catalase and glutathione in post thawed samples seminal plasma were determined. The present results indicated that adding 300 µg of methanolic Moringa oleifera leaves extract to each 1 ml of citrate-egg yolk extender as an alternative to penicillin and streptomycin can improve quality of Friesian bull semen during different preservation stages.
