Effect of bacterial endotoxin and interleukin-1 beta on hippocampal serotonergic neurotransmission, behavioral activity, and free corticosterone levels: an in vivo microdialysis study

Abstract: In this study the effect of immune system stimulation and intracerebroventricular (i.c.v.) administration of interleukin-1 beta (IL-1 beta) on hippocampal serotonergic neurotransmission, behavioral activity, and the hypothalamic-pituitary-adrenocortical (HPA) axis is described. An in vivo microdialysis method was used to measure hippocampal extracellular concentrations of serotonin (5-HT) and its metabolite 5-hydroxyindoleacetic acid (5-HIAA) in conscious, freely moving rats. In addition, we established a meth… Show more

“…Other studies have indicated elevations of 5-HT release from the hippocampus [147,148], as well as NE, MHPG, 5-HT and 5-HIAA in the preoptic area [149]. In the nucleus accumbens, LPS injection (100 μg ip) increased DA and 5-HIAA [150].…”

“…IL-1␤ adminstration in the CNS has been reported to produce 'stress-like' effects on behavior, food intake, monoamine neurotransmitters, hypothalamic pituitary adrenal axis activity and immune function. [21][22][23][24][25][26][27][28] IL−1 is a potent stimulus to corticotropinreleasing hormone (CRH) production, resulting in activation of the hypothalamic-pituitary-adrenal (HPA) axis. 29,30 CRH hypersecretion is a well replicated finding in depression.…”

“…Systemic administration of the bacterial endotoxin lipopolysaccharide (LPS) is a widely used and accepted model to emulate immune activation and subsequent release of immunoregulatory, cytotoxic and inflammatory cytokines secondary to gram-negative bacterial infections (Dinarello 1984;Andersson et al 1992). Administration of LPS or pro-inflammatory cytokines causes pronounced centrally mediated effects, such as changes in body temperature (Linthorst et al 1994;Pauli et al 1998), sleep (Pollmächer et al 1993, neuroendocrine function (Besedovsky et al 1986;Berkenbosch et al 1987;Cambronero et al 1992;Linthorst et al 1994Linthorst et al , 1995Molina-Holgado et al 1996;Pauli et al 1998) and behavior (del Cerro andBorrell 1990;Kent et al 1992;Linthorst et al 1995;Anisman et al 1998;Hanisch et al 1997;Pauli et al 1998;Zalcman et al 1998).To investigate the putative effect of a prenatal immune challenge on sensorimotor gating, a new experimental design was developed consisting in the administration of LPS to pregnant rats followed by evaluation of PPI in the adult offspring. We show that prenatal LPS treatment disrupts sensorimotor gating later in life.…”
