Effect of Bavistin and Adenine Sulphate on In vitro Shoot Multiplication of Picrorhiza scrophulariiflora

Abstract: An alternative protocol for in vitro propagation of Picrorhiza scrophulariiflora is described using bavistin and adenine sulphate. The explants differentiated into multiple shoot buds on MS supplemented with various concentrations of bavistin and adenine sulphate ranging from 0 -400 mg/l either alone or in combination. Maximum number of multiple shoots were obtained on MS containing the combination of bavistin (100 mg/l) and adenine sulphate (100 mg/l). In this combination as high as 28 shoots per explant was … Show more

“…Jia et al 1999). P. scrophulariiflora contains an additional glycosides which are absent in Picrorhiza kurroa (Li et al 1998, Smith et al 2000 and total picroside content is much higher than in P. kurroa (Bantawa et al 2009b). Conventional propagation methods through seed and root tubers for cultivation of this plant are too slow.…”

“…Our group have established a reproducible micropropagation protocols (Bantawa et al 2009b(Bantawa et al , 2010 and somatic embryogenesis (Bantawa 2009) of P. scrophulariiflora. However, there is no report on organogenesis of this plant, , which is the prerequisite for any genetic manipulation and for conservation of this endangered medicinal plant.…”

“…The beneficial effects of ethylene inhibitors on organogenesis for plant regeneration have been widely reported [12][13][14]. The influence of Bavistin and silver thiosulphate has been well documented in many medicinal plants such as Mentha piperita [15], Stevia rebaudiana [16] and effect of Bavistin with adenine sulphate has been studied in Picrorhiza scrophulariiflora [17].…”

“…phulariiflora [8]. Since studies on in vitro plant regeneration of this species using different antimicrobial agents and ethylene inhibitor STS are almost limited, hence in the present study, we report a protocol on the effect of different concentrations of bavistin; cefotoxime and kanamycin along with STS were examined.…”
