Effect of Bovine Serum Albumin on Mitochondrial Respiration in the Brain and Liver of Mice and Rats

Abstract: We studied the effect of BSA (in the isolation medium) on the oxidation rate of succinate, glutamate, pyruvate, and α-ketoglutarate by mitochondria of the brain and liver from C57Bl/6g mice and Taconic Sprague Dawley rats. BSA had no effect on liver mitochondrial respiration, but increased oxidation of substrates (particularly of succinate) in brain mitochondria. Therefore, the major effect of BSA on brain mitochondria is manifested in activation of SDH. The improvement of mitochondrial properties in the brain… Show more

“…Rotenone, however, inhibits succinate-stimulated ROS production by RET. It has been suggested that RET is physiologically regulated by several mechanisms, 1) by control of succinate dehydrogenase activity in the citric acid cycle (39), 2) by fatty acids (40), 3) and by mitochondrial membrane potential (35). Thus RET is not an "artifact" and rotenone supplementation may produce an artificially improved mitochondrial functions.…”

“…The degree of this intrinsic SDH inhibition varies strongly between the species. In other words, it depends on the metabolic phenotype of the BM [62, 126]. …”

“…Therefore, most researchers traditionally isolate BM and SCM in the presence of defatted BSA [126], which significantly improved oxidation of succinate (Figure 7(b)). Figure 7(b) shows, however, that even with the BSA-BM oxidizing succinate, it required more than a minute for the membrane potential to reach the maximum.…”

“…However, absolute rates respiration at the State 4 and State 3, are also very important for understanding the mitochondrial energetics. Generally, in mitochondria from normal tissues isolated in the presence of bovine serum albumin, the RCR values are higher with the NAD-dependent substrates than with succinate [51]. …”
