Effect of clonidine on haemodynamic responses to endotracheal intubation and on gastric acidity

Abstract: Sixty-three patients (ASA 1-2), scheduled for elective surgery under general anaesthesia, were randomly given either oral clonidine (225-375 micrograms) + diazepam (5 15 mg), cimetidine (300 mg the night before and 300 mg in the morning) + diazepam or only diazepam for premedication. Anaesthesia was induced with thiopentone and maintained with N2O + O2 (70:30), enflurane and fentanyl. Vecuronium bromide was used as a muscle relaxant. The sleep dose of thiopentone was significantly smaller in the patients pretr… Show more

“…Bloor & Flacke [129] also reported an anaesthetic-sparing effect in dogs, the administration of clonidine decreasing the MAC of halothane by a maximum of 50%. In addition the dose of thiopentone required for induction of anaesthesia is reduced by clonidine premedication [130].…”

“…Similarly, preanesthetic medication with 225-375~tg oral clonidine (approximately 3.5-4.7,ag.kg -1) plus 5-15 mg diazepam reduced thiopental requirements by approximately 20% in adult humans [8]. Our present results that 5.0 and 2.5 ~tg'kg -1 oral clonidine reduced thiamylal requirements by 23% and 18%, respectively, agree with those of Orko et al [8]. However, we observed a smaller reduction in barbiturate requirements than had been reported previously Values are mean +_ SE, with ranges in parentheses.…”

“…Likewise, intravenous clonidine [7] and oral clonidine with diazepam [8] reduced thiopental requirement for induction of anesthesia in adults. Recently, it has been shown that preanesthetic administration of 2 ~g-kg -1 or 4 ag.kg -~ oral clonidine reduce thiamylal requirement in children in a dose-dependent manner by approximately 17% and 37%, respectively [9].…”
