Effect of Crocetin on Blood Pressure Restoration and Synthesis of Inflammatory Mediators in Heart After Hemorrhagic Shock in Anesthetized Rats

Abstract: Crocetin, a constituent of saffron, has been shown not only to prevent reactive oxygen species-induced hepatotoxicity and genotoxicity but also to increase whole-body oxygen consumption and survival. The present study was to determine whether crocetin has beneficial effects on cardiac injury caused by hemorrhagic shock and resuscitation in rats. Anesthetized rats were bled to reduce mean arterial pressure (MAP) to 35 +/- 5 mmHg for 60 min and then resuscitated with their withdrawn shed blood and isotonic sodiu… Show more

“…Mean arterial pressure was maintained at 35 (SD, 5 mmHg) for 30 min by further withdrawal or reinjection of blood as required. In the crocetin group at the end of this period, crocetin was given as a bolus of 50 mg/kg suspended in 0.5% CMC-Na though the duodenal catheter (9). Animals in the sham group and the control group were given an equivalent volume of 0.5% CMC-Na.…”

“…15 Absorbance was determined at 660 nm on a StatFax: 2100 spectrophotometer (Awareness Technology, Palm City, FL, USA). Serum MDA content was measured using the thiobarbituric acid (TBA) method, based on the formation of a red complex when MDA reacts with TBA.…”

“…The addition of molybdenum acid ammonium produces phosphomolybdic acid, which can be further reduced to molybdenum blue. Enzymatic activity was calculated from the amount of inorganic phosphorus produced, and absorbance detection was set at 660 nm [20]. SOD levels were determined by the xanthine oxidase method, as the xanthine and xanthine oxidase reaction systems produce superoxide anion, which oxidize hydroxylamine to form nitrite.…”
