Effect of Cytochrome c on the Generation and Elimination of O2 and H2O2 in Mitochondria

Abstract: The primary recognized function of cytochrome c is to act as an electron carrier transferring electrons from complex III to complex IV in the respiratory chain of mitochondria. Recent studies on cell apoptosis reveal that cytochrome c is responsible for the programmed cell death when it is released from mitochondria to cytoplasm.

“…The primary source of ROS in sperm is the respiratory chain of the mitochondrion (5)(6)(7)(8). The main ROS form in sperm is hydrogen peroxide (H 2 O 2 ), the concentration of which in sperm has not been measured (2)(3)(4), whereas in liver cells, the H 2 O 2 concentration (10 Ϫ7 to 10 Ϫ9 mol͞liter) is known to be 100-to 10,000-fold higher than that of superoxide (O 2 ⅐Ϫ ) (Ϸ10 Ϫ11 mol͞ liter) (6,9,10). O 2 ⅐Ϫ , as the precursor of H 2 O 2 and a short-lived ion, can be rapidly converted to H 2 O 2 by several pathways and does not accumulate in the cell (10,11).…”

“…Complex I and III of the ETC have been identified as the two principal sites of superoxide anion (O 2 -) generation [52]. While much of the O 2 -is released from complex III to either side of the inner membrane, the iron-sulfur cluster N 2 of complex I appears to be the main site of O 2 -release to the matrix [53]. The O 2 -so formed is disposed of by several pathways.…”
