Effect of estradiol during culture of bovine oocyte–granulosa cell complexes on the mitochondrial DNA copies of oocytes and telomere length of granulosa cells

Abstract: During the development of oocytes from early antral follicles (EAFs) to antral follicles (AFs), the mitochondrial DNA copy number (Mt DNA number) increases, and granulosa cells markedly proliferate. This study examined the effect of supplementation of culture medium with estradiol-17β (E2) on the in vitro growth of oocytes, and increases in the Mt DNA number, and telomere length during the in vitro culture of oocytes derived from EAFs (0.4-0.7 mm in diameter). The E2 supplementation improved antrum formation a… Show more

“…Thus, it is plausible that mtDNA number is controlled by both biosynthesis and degradation. In addition, our recent study shows that mtDNA number in oocytes increases during in vitro oocyte growth from the early antral follicle stage to the antral follicle stage and that the increase ratio varies to a great extent and depends on the individual donors [30]. In agreement with this result, in the present study, the increase ratio for mtDNA number also varied ranging from 0.15 to 6.26.…”

“…Based on these results, we concluded that the stimulatory effect of Res on mitochondrial biosynthesis depends on the individual conditions of donor cows and that there may be additional unknown factors in oocytes contributing to mitochondrial biosynthesis. In agreement with these results, when oocytes of early antral follicles were cultured and the kinetics of Mt numbers in oocyte were examined, the ration of increase in Mt number in oocytes varied substantially among donor cows [29]. We observed a similar trend in our results for MMP.…”

“…Mitochondrial number can be predicted by quantifying the mitochondrial DNA copy number (Mt number) using real time-PCR, based on reports that mitochondria in oocytes typically contain one copy of the mitochondrial genome [4]. The Mt-number in bovine oocytes increases during the growth of oocytes from early antral follicles to antral follicles in vivo and in vitro [5], [6], and the number remains the same or increases further during oocyte maturation [5], [7], [8]. However, little is known about how culture conditions affect Mt number in oocytes.…”
