Effect of Huaier On the Proliferation of Mesangial Cells in Anti-Thy-1 Nephritis

Abstract: Background/Aims: To determine whether an aqueous extract of Trametes robiniophila Murr. (Huaier) suppresses anti-Thy-1 mesangial proliferative glomerulonephritis (MsPGN) in vivo and platelet-derived growth factor (PDGF)-BB–induced mesangial cell proliferation in vitro. Methods: Male Wistar rats were randomly categorized into 5 groups: Sham, Thy-1, and 3 Huaier-treated groups (low, medium, and high dose). Two weeks after treatment, urinary proteins were quantified and renal pathological changes were examined. M… Show more

“…CASY cell analysis confirmed that Huaier effectively inhibited the proliferation of Hacat cells in a statistically significant time- and concentration-dependent manner (P <0.01). This is similar to many other studies, which have indicated that Hacat can inhibit the proliferation of tumor cells: previous experiments have confirmed that Huaier effectively inhibited the proliferation of melanoma, breast cancer, colon cancer, liver cancer, prostate cancer, stomach cancer, fibrosarcoma, lung cancer, and many other tumor cells [ 3 , 8 14 ]; in addition to tumor cells, previous studies have confirmed that Huaier inhibited the proliferation of some nontumor cells, such as mesangial cells and nodular sclerosis cells [ 15 , 16 ]. This study confirmed that Huaier effectively inhibited the excessive proliferation and accumulation of keratinocytes, which demonstrated an important link to the pathogenesis of psoriasis.…”

“…According to the research of Bai et al, PDGF-B is involved in the development of mesangial cells and overexpression of glomerular PDGF-B has an important effect on MsPGN disease. The research indicates that HQH can reduce the stimulation of PDGF-B, decrease the urinary protein, prolong G2 phase, and inhibit cell proliferation by regulating cyclin, so as to reduce the proliferation of anti-Thy-1 MsPGN [15]. As a result, HQH inhibits mesangial proliferation in MsPGN, promising a possible therapy in its clinical treatment.…”

“…Studies of Li et al have shown that IgG is significantly higher after treatment with HQH than before treatment and CD3; CD4 are significantly higher than before treatment. Therefore, HQH can regulate the immune function and reduce infection in children with IgA nephropathy, indicating a quickly and effectively to treat it 15 . IgA can be improved by the antiproliferation and immunomodulatory properties of HQH.…”

“…Currently, mesangial cells are recognized as the major cells to secrete ECM [ 5 ]. Therefore, inhibiting the proliferation of mesangial cells and the accumulation of ECM can be applied as a practical approach for treating or relieving DN [ 6 ]. However, the underlying mechanism is still unsatisfactory, and there is no effective therapeutic treatment drug.…”
