Medium containing Fetal Bovine Serum (FBS) provides a supportive environment for isolation and expansion of mesenchymal stromal/stem cells (MSCs); however, the inherent variability of FBS may contribute to inconsistencies in cell growth and yield between batches of stem cell products. For this reason, we set out to develop a serum-free medium capable of supporting the in vitro expansion of MSCs. First a naïve serum-free medium was formulated by Sato’s approach. Once it was established that the naïve serum-free medium supported the expansion of canine adipose-derived MSCs (Ad-MSCs), the serum-free medium was optimized by addition of growth factors. Combinations of growth factors were chosen and compared by their effect on cell proliferation and colony formation. Growth characteristics of canine adipose-derived MSCs cultured in the serum-free medium were comparable to those cultured in standard FBS containing medium. In addition, cell surface marker expression and differentiation potential of serum-free and FBS-based cultures were also comparable. However, a commercial serum-free medium developed for human MSC culture did not support growth of canine Ad-MSCs. In summary, canine Ad-MSCs isolated and cultured in serum-free medium retained the basic characteristics of MSCs cultured in FBS containing medium.

Previous work, almost four decades ago, showed that hydrocortisone (HC) treatment reduces the number of skeletogenic condensations that give rise to the scleral ossicles in the chicken eye. The scleral ossicles are a ring of overlapping intramembranous bones, the sclerotic ring, and are present in most reptiles, including birds. The scleral condensations that give rise to the scleral ossicles are induced by a series of overlying thickenings (or papillae) of the conjunctival epithelium. Here, we further explore the effects of altering the dosage and timing of HC treatment on the morphology and number of skeletogenic condensations and conjunctival papillae. We show that high doses can completely obliterate the entire sclerotic ring. Significantly, the reduction in papillae number we observed was less extreme than that of the scleral condensations, indicating that additional factors contribute to the observed skeletogenic condensation loss. Via immunohistochemical analyses, we show that HC treatment alters the spatial expression pattern of several extracellular matrix components (tenascin-C, decorin and procollagen I) and also alters the vasculature network within the sclera. This research provides important insights into understanding the role of the scleral tissue components in ossicle development within the vertebrate eye.

Engineering clinically-relevant musculoskeletal tissues at a human scale is a considerable challenge.Developmentally-inspired scaffold-free approaches for engineering cartilage tissues have shown great promise in recent years, enabling the generation of highly biomimetic tissues. Despite the relative success of these approaches, the absence of a supporting scaffold or hydrogel creates challenges in the development of large scale tissues. Combining numerous scaled-down tissue units (herein termed microtissues) into a larger macrotissue represents a promising strategy to address this challenge. The overall success of such approaches, however, relies on the development of strategies to support the robust and consistent chondrogenic differentiation of clinically relevant cell sources such as mesenchymal stem/stromal cells (MSCs) within microwell arrays to biofabricate numerous microtissues rich in cartilage-specific extracellular matrix components. In this paper, we first describe a simple method to manufacture cartilage microtissues at various scales using novel microwell array stamps. This system allows the rapid and reliable generation of cartilage microtissues, and can be used as a platform to study microtissue phenotype and development. Based on the unexpected discovery that Endothelial Growth Medium (EGM) enhanced MSC aggregation and chondrogenic capacity within the microwell arrays, this work also sought to identify soluble factors within the media capable of supporting robust differentiation using heterogeneous MSC populations. Hydrocortisone was found to be the key factor within EGM that enhanced the chondrogenic capacity of MSCs within these microwell arrays. This strategy represents a promising means of generating large numbers of highquality, scaffold-free cartilage microtissues for diverse biofabrication applications.
