Effect of Hydrocortisone on Multipotent Human Mesenchymal Stromal Cells

Abstract: We studied the effect of natural glucocorticosteroid hydrocortisone on total cell production, cloning efficiency, and expression of genes important for the function of mesenchymal stromal cells. Addition of hydrocortisone to the culture medium reduces the total cell yield by 2 times and significantly increased cloning efficiency by 2-3 times; this effect was more pronounced in multipotent mesenchymal stromal cells obtained from female donors. Hydrocortisone had no effect on the expression of immunomodulatory f… Show more

“…FBS contains adrenocortical hormones such as hydrocortisone (HCN), which is important for adhesion and growth of human MSCs [29, 30]. Typical concentrations of HCN ranged from 100 nM to 1 μM [30]. In this study, the serum-free medium was supplemented with 50 to 100 nM of HCN (Sigma, St. Louis, MO).…”

“…Procollagen I is the precursor for collagen I, which is the main extracellular component of bone (Peltonen et al 1985;Mackie et al 1987;Canty-Laird et al 2012). Glucocorticoids, such as HC, can reduce osteoblast proliferation, increase apoptosis of osteoblasts, and increase osteoclast activity (Weinstein et al 1998;Silvestrini et al 2000;Canalis & Delany, 2002;Sch€ acke et al 2002;O'Brien et al 2004;Shipunova et al 2013). In this study, HC treatment resulted in reduced procollagen I expression in the sclera, which may have been insufficient to support osteoblast differentiation.…”

“…As such, it is often included in serum-free medium formulations. Additionally, hydrocortisone has been used to 'activate' multipotent MSCs for adipogenic differentiation, while its addition during passaging helps to preserve the self-maintenance capacity of MSCs [56]. In the context of chondrogenesis, hydrocortisone supplementation in 3D culture with human chondrocytes has been shown to optimise ECM metabolism.…”
