Effect of Lipid-Hydroperoxide-Induced Oxidative Stress on Vitamin E, Ascorbate and Glutathione in the Rabbit Retina

Abstract: Background: It is possible that oxidative stress causes several retinal diseases. However, the natural biogenic role of antioxidants in the retina is not clear. Purpose: This study investigates the change in concentration of vitamin E (VE), ascorbate and glutathione (GSH) in the retina following vitreous injection of 600 µg 18:2 linoleic acid hydroperoxide (LHP) in male New Zealand rabbits. Method: LHP was injected above the retinal surface. The animals were sacrificed and the eyes enucleated before LHP inject… Show more

“…12 The drop of these antioxidants could reveal the switch from their reduced form to their oxidized form. Kamegawa et al 26 similarly reported a fall of the reduced form of several antioxidants, such as glutathione, ascorbate, and vitamin E in rabbit retinas a few hours after a pro-oxidative molecule (lipid hydroperoxide) was injected intravitreally. The present study showed that the antioxidant decrease is an early event after PVL treatment, because glutathione, taurine, and ascorbate dropped strongly only two hours after incubation with the toxin.…”

“…The structure and function of the retina rely largely on fatty acid composition [27] . According to evidence from epidemiological studies and animal experiments, the fatty acid composition of the retina is in uenced by diet [28][29][30] . The ndings in this study seem to suggest that nutrient intake and metabolic organ function may both affect ocular health through metabolism, providing new ideas for intervention strategies.…”

“…Additionally, the groups treated with puerarin exhibited significantly higher AOC compared with the untreated diabetic group. GSH is an important antioxidant agent in the retina and serves a role similar to vitamins A, E and C, and selenium in the defense against cataract formation (46). In the present study, the levels of GSH in puerarin-treated rats were higher compared with untreated diabetic rats.…”
