This paper presents a review of studies related with the production of specialty lipids by the yeast Yarrowia lipolytica during growth on various fatty agro-industrial residues utilized as substrates. In a first approach, the potential for growth on stearin (a low-cost industrial derivative of tallow composed of saturated free fatty acids) was assessed; significant biomass production was obtained, accompanied by notable intracellular accumulation of lipid which occurred as a primary anabolic activity regardless of the extracellular nitrogen availability in the medium. Y. lipolytica consumed its own storage lipids in the stationary growth phase. The secretion of extracellular lipase in non-negligible quantities was also observed in cultures performed on stearin. Although Y. lipolytica tended to increase the intracellular level of stearic acid, its reserve lipid could be used as a cocoa butter substitute, provided that an oleic acid donor, such as chemically hydrolyzed rapeseed oil, is used as co-substrate together with stearin. Accordingly, predetermined lipids with composition similarities to cocoa butter were synthesized using low-cost substrates. Other strategies related with the production of microbial cocoa butter substitutes are also presented and comprehensively discussed.

An extracellular acidic lipase producing fungi, screened and isolated from Palm Oil Mill Effluent (POME) dump sites, Pedavegi, West Godavari Dist, A.P., was found to be Emericella nidulans NFCCI 3643 by ribotyping of 28S rRNA gene. BLAST search following analysis of phylogenetic tree revealed that its closest phylogenetic neighbor is Emericella fruticulosa strain CZ032. Optimization of process variables for the enhanced lipase production from Emericella nidulans NFCCI 3643 was carried out using traditional OFAT (One Factor At a Time) method and found that the best production medium for highest lipase yield was Lipase Production Medium (LPM) 3 containing 1.5% olive oil as sole carbon source, 1% (NH 4 ) 2 SO 4 as nitrogen source, 0.5% gum arabic as surfactant, pH 6 at 30°C and 150 rpm.

Three kinds of matrices (calcium alginate, gelatin, and PVA) were employed as supports to immobilize lipases from Y. lipolytica KKP 379 via physical adsorption. The stability of biocatalysts (free and immobilized) was evaluated by measuring the enzyme activity before and after treatment with the method based on the hydrolysis of p-nitrophenyl laurate. Two fractions of enzymes were immobilized: cell-bound (yeast biomass) and extracellular (supernatant). The yield of immobilization and catalytic properties of immobilized lipases were investigated. Satisfactory results for lipolytic activity and biocatalyst stability were obtained for cell-bound enzymes immobilized in alginate (0.38 U g -1 d.m.) and crosslinked gelatin (0.18 U g -1 d.m.). Immobilization of the supernatant was successful only on the alginate (0.026 U g -1 d.m.). After lyophilization, no significant difference was noticed between treated and untreated biocatalysts. Lyophilized catalysts were successfully immobilized in all three matrices, but the process reduced their lipolytic activity probably due to an insufficient amount of water in the reaction solution.
