Effect of lipoarabinomannan from Mycobacterium avium subsp avium in Freund’s incomplete adjuvant on the immune response of cattle

Abstract: The aim of the present study was to determine whether lipoarabinomannan (LAM), in combination with Freund's incomplete adjuvant (FIA), was able to improve cell-mediated and antibody-mediated immune responses against ovalbumin (OVA) in cattle. Twenty-three calves were assigned to four treatment groups, which were subcutaneously immunized with either OVA plus FIA, OVA plus FIA and LAM from Mycobacterium avium subsp avium, FIA plus LAM, or FIA alone. Lymphoproliferation, IFN-γ production and cell subpopulations o… Show more

“…The first injection was emulsified with Freund’s complete adjuvant (Sigma), and the remaining two injections were emulsified with Freund′s incomplete adjuvant (Sigma). Freund’s complete adjuvant is considered to be the gold standard adjuvant for immunization [ 22 ]. The Freund’s complete adjuvant has been commonly used to induce antibody production in animal models [ 23 ].…”

“…Stimulation of cattle peripheral blood mononuclear cells (PBMCs) with LAM isolated from M. avium along with Freund’s incomplete adjuvant (FIA) induces secretion of IFN-γ. The immune stimulatory function of LAM is exerted through a MyD88 directed CARD9/NF-κB pathway and NLRP3 inflammasome activation, suggesting that M. avium LAM could be used as a potential vaccine candidate against bovine tuberculosis (158). PGN of bacteria is not a potent immune adjuvant; however, peptidoglycan monomer (PGM) induces appropriate immune responses without any pyrogenic and toxic effects (159).…”
