Effect of Matrigel on the Osteogenic Potential of Canine Adipose Tissue-Derived Mesenchymal Stem Cells

Abstract: ABSTRACT. Adipose tissue-derived mesenchymal stem cells (Ad-MSCs) are a promising source of cells for bone tissue engineering. Matrigel is a basement membrane extract containing multiple extracellular components. This mixture may promote the osteogenic differentiation of MSCs and provide a more appropriate microenvironment for transplanted cells. Here, we investigated the effect of Matrigel on the osteogenic potential of Ad-MSCs. Canine Ad-MSCs were cultured in 2D and 3D matrices and implanted into subcutaneou… Show more

“…For example, breast epithelial cells ceased proliferating, formed acinar-like structures, and showed a large increase in the production of milk proteins when grown on BME/Matrigel. Similarly, when grown on BME/Matrigel, bone cells ceased proliferation and formed canaliculi [11][12][13][14][15]. Bone cell differentiation studies also showed the importance of specific growth factors when the growth factor reduced BME/Matrigel was supplemented with various growth factors.…”

“…fibrinogen or hyaluronic acid. Notably, these scaffolds are mainly used for the regenerative cell therapy of bone fractures [13,14]. In some cases MSCs were applied only once and shortly after the injury [15].…”

“…In addition, the easy and inexpensive production of this scaffold makes it more suitable for the repair of bone defects than the previous artificial scaffold materials. Different types of cells have been used for bone tissue engineering, including bone marrow cells, periosteal cells, osteoblasts derived directly from bone, and MSCs derived from various tissues [5,14,15,21]. Ideal cells seeded for bone engineering should be able to proliferate quickly, function like osteoblast, be non-immunologic when implanted in vivo, and be biocompatible with scaffold materials [2,22].…”

“…Previously, we isolated and characterized the MSCs derived from the adipose tissue of dogs [4]. In addition, we have demonstrated that canine adipose tissue-derived MSCs (Ad-MSCs) mixed with beta-tricalcium phosphate and Matrigel have osteogenic potential in ectopic implantation [5]. Ad-MSCs have a good ability to differentiate into osteoblasts, and clear advantages including easy and repeatable access to subcutaneous adipose tissue; a simple isolation procedure; abundance in supply; and a relatively noninvasive harvesting procedure [6].…”
