Effect of Meta-topolin on in vitro propagation of Pelargonium × hortorum and Pelargonium × hederaefolium cultivars

Abstract: The aim of this study was to develop an efficient in vitro propagation method of Pelargonium plant (seven cultivars) using axillary buds and shoot tips as explants. The influence of the time period in which the explants were collected and the influence of the growth regulators (BAP, meta-topolin, IBA) were studied. The explants taken in April had a higher regeneration ability than those isolated in July and September. The most efficient regeneration and axillary multiplication were achieved on the medium supple… Show more

“…Susceptibility to early senescence greatly depends on plant species. Our previous [2,3] and this study showed significant differences among Pelargonium genotypes in their susceptibility to leaf yellowing. Early senescence is widely influenced by stress conditions [18].…”

“…The shoots were subcultured at three week intervals into a fresh medium in an Erlenmeyer flask and kept at a temperature of 23°C, under a 16 h photoperiod provided by cool-white fluorescent lamps at 40 μmol m ) without growth regulators (control) or supplemented with meta-topolin (mT, 2.07 μM) alone or together with a precursor of ethylene biosynthesis -1 aminocyclopropane-1-carboxylic acid (ACC, 9.9 μM), an inhibitor of ethylene synthesis -aminoethoxyvinylglycine (AVG, 5.1 μM), or an inhibitor of ethylene action -AgNO 3 (5.9 μM). The concentration of growth regulators was determined on the basis of previous experiments (Wojtania, 2010;Wojtania and Węgrzynowicz-Lesiak, 2012).…”

“…Articles published so far have enumerated the following cultivars of Pelargonium × hortorum : "Bargpalais", "Holywood Red", "Grand Prix", "Jazz Rocky Mountain", "Orbit White", "Scarlet Orbit", "Red Elite", "Ringo Rose", "Ringo Salmon", "White Rocky Mountain", and Pelargonium × hederaefolium ! "Beach", "Bonete", "Luna", "Sofie Cascade", and such like (Desiltes et al, 1993;Murthy et al, 1996;Mithila et al, 2001;Wojtania and Gabryszewska, 2001;Wojtania, 2010;Wojtania et al, 2004;Madden et al, 2005), with petals having many colors, different foliage types and growth habits. In our experiments, we focused on the elaboration of a convenient propagation medium for Pelargonium × domesticum.…”
