Effect of ouabain on myocardial remodeling in rats

Abstract: The aim of this study was to investigate the effect of ouabain (EO) on myocardial remodeling. Twenty-two adult male Sprague-Dawley rats were randomly divided into two groups: the rats in the EO group (n=12) were intraperitoneally injected with EO daily and those in the control group (n=10) were injected with physiological saline daily. After 8 weeks the rats were sacrificed. The ultrastructural changes in the myocardium were observed. The expression levels of voltage-gated potassium channel 4.2 (KV4.2) were de… Show more

“…The doses infused were estimated to increase the plasma ouabain concentration by 0.5–1.0 nmol/l above the normal physiological level (approximately 0.3–0.9 nmol/L) [ 29 ], thus leading to an ouabain concentration similar to that in pathological conditions such as hypertension and heart failure (approximately 0.9–1.8 nmol/L) [ 30 ]. Intraperitoneal injection of ouabain daily for consecutive eight weeks has been proven to effectively increase blood pressure in SD rats [ 21 , 22 ].…”

“…Intracerebroventricular injection of antibody Fab fragments against ouabain could effectively inhibit excess sympathetic activation and prevent the elevation of blood pressure in Dahl salt-sensitive hypertensive rats [ 17 ]. Hypertensive animal models can be established through intraperitoneal injection of ouabain [ 18 22 ].…”

“…Several lines of evidence suggest that endogenous ouabain could be involved in the pathogenesis of essential hypertension. It affects the function of all cells by binding to Na + /K + –ATPase (Meira et al , 2014; Miles et al , 2013) and associated with increased sympathetic activity (Pavlov et al , 2005; Ren et al , 2013). Moreover, it induces activation of the brain renin-angiotensin-aldosterone system and promotes increases in the activity of endothelial angiotensin-converting enzyme (ACE) and the local synthesis of angiotensin II (Ketchem et al , 2016; Padilha et al , 2011).…”

“…Changes in ion currents, including Ca 2+ and K + , form the basis for early electrical remodeling in AF (8,9). Calcium entry from the extracellular space through LTCCs and the resultant intracellular Ca 2+ elevation (calcium overload) was demonstrated to be crucial in the regulation of atrial frequency-dependent action potential duration (APD) and effective refractory period (ERP) (10). The transient outward K + current mediated by the potassium channel Kv4.3 contributes to early repolarization (11).…”
