Effect of Prednisone Treatment on the Human Immune Response to Keyhole Limpet Hemocyanin

Abstract: The immune responses to KLH injections were investigated in 12 children with asthma, 6 of whom received prednisone for control of asthma. Two injections of 320 μgN KLH were given 14 days apart. Antibodies were investigated by hemagglutination titration, radioactive antigen-binding techniques and skin testing. The results suggested that therapeutic doses of prednisone, given on alternate days, may have enhanced the primary γG- and γM-antibody responses, but had no effect on antibody levels formed after the boos… Show more

“…Recently, it has been reported [17] that (alternate day) therapy with prednisone was associated with an en hanced primary response (at 2 weeks) in asthmatic children immunized with keyhole limpet hcmocyanin (KLH). Since other asthmatic children not receiving prednisone showed a smaller antibody response to KLH at 2 weeks, the results suggest that corticosteroids may have an augmenting effect on the primary immune response, at least in this group of patients.…”

“…When corticosteroids inhibit a suppressor infl uence, antibody production may actually increase. Enhancement of antibody production by cortico steroid administration has been reported in animals (84) and man (85). The addition of pharmacologic or physiologic concentrations of corticosteroid to an in vitro system of antibody production by human B lymphocytes inhibited the negative effect and produced an enhanced antibody response (86).…”

“…More recent findings suggest that high dose administration of steroid results in modest decreases in serum immunoglobulins, particularly IgG and IgA, which may persist for periods of several months after treatment is stopped (5,6). Specific antibody synthesis in man, however, has not been shown to be suppressed by corticosteroids, and in one study, enhancement was demonstrated (7). In young chickens, treatment with cortisone resulted in the mobilization of B lymphocytes from the Bursa and enhanced the differentiation into mature immunoglobulin secreting plasma cells in the peripheral lymphoid tissues (8).…”
