Effect of presence of corpora lutea on cumulus expansion of in vitro matured bovine oocytes selected by trypan blue and brilliant cresyl blue tests

Abstract: Trypan Blue (TB) and Brilliant Cresyl Blue (BCB) are used to select and determine viability and competence of oocytes, however, the effects of corpora lutea on cumulus expansion during in vitro maturation are still undetermined. Cumulus-oocytes complexes (COCs) from ovaries with ipsilateral (ICL), contralateral (CCL) and without corpora lutea in either ovary (OCL) were selected by visual examination, TB and BCB staining, and matured in vitro to evaluate cumulus expansion. The overall percentage of visually sel… Show more

“…Some studies have shown that the corpus luteum in an ovary enhances the quality and developmental competence of oocytes derived from that ovary (Reis et al, 2006;Moreno et al, 1993;Penitente-Filhe et al, 2015). However, our study findings contrast those of other studies (Shabankareh et al, 2015;Quezada-Casasola et al, 2018), as well as subsequent embryonic development (Gonzalez-Bulnes et al, 2005;Manjunatha et al, 2008;Boediono et al, 1995;Pirestani et al, 2011). Additionally, several studies have reported no effects (de W it et al, 2000;Sungulle, 2008).…”

“…Fertilization success in in vitro embryo production is typically high (~90%), but a significant proportion of embryos fail to develop to blastocysts (Carter et al, 2008;Lonergan et al, 2016). The environment of in vitro culture to which the embryo is exposed is crucial in determining the rate of embryos to develop further stages (Quezada-Casasola et al, 2018;Sen and Kuran, 2018). Most studies have agreed that an elevated concentration of circulating progesterone in vivo is associated with an increase in pregnancy rate in cattle (Stronge et al, 2005;McNeill et al, 2006).…”

“…The presence of a corpus luteum (CL) and/or progesterone in mice affects estradiol concentration, which is required for the acquisition and maintenance of CCs to undergo expansion [42]. In bovines, the presence of a CL in the ovary exerts a negative effect on the expansion of CCs, suggesting that the CL receives the highest rate of blood flow out of all the other ovarian tissues [43].…”
