Salinity is an important problem all over the world. The destructive effect of salinity is observed from the seed germination stage. In this study, it was aimed to determine the effect of salinity on seed germination of the medically important Satureja thymbra L., whether pre-treatments are a factor in breaking the salinity stress, and to determine the level of salinity tolerance of this species. In the research, firstly, the seeds were exposed to two pretreatments (80°C (5 minutes) + 10 ppm GA3 (24 hours), 80°C (5 minutes) + 100 ppm GA3 (24 hours)) and then 8 different NaCl concentrations (0.1 g/l, 1 g/l, 2.5 g/l, 5 g/l, 7.5 g/l,10 g/l, 15 g/l and 30 g/l) were tried. Germination seeds were counted every day and the effects of salinity on germination characteristics were investigated. The highest germination percentage (90%) was obtained at 0.1 g/l NaCl after 80°C (5 min.) + 100 ppm GA3 (24 h.) pretreatment. The results showed that the effect of salinity was significant on germination parameters in p < 0.05. Obtained results showed that the highest NaCl concentration at which Satureja thymbra seed could germinate was 10 g/l.

Quinoa (Chenopodium quinoa Willd.) is a facultative halophyte renowned for its importance in enhancing food security, and it supports forage production across diverse climatic regions. The objective of this study is to examine the impacts of multiple pre-treatment methods on C. quinoa seed (Titicaca cultivar) germination parameters, identify the optimum pre-treatment to diminish the consequence of salinity, and promote the productivity of this crop, especially in marginal environments. For this purpose, a spectrum of sodium chloride (NaCl) concentrations spanning from 0 to 500 mM and gibberellic acid (GA3) concentrations ranging from 0 to 300 ppm were tested, and mechanical scarification (MS) was carried out. The effect of a combination of these pretreatment NaCl/GA3 and NaCl/MS on the germination parameters of C. quinoa seed was also investigated. The results showed that the total germination, vigor index, and germination index decreased progressively with an increase in salinity. Hence, salinity exhibited a notable influence on most germination parameters. Moreover, seeds scarified with 500 mM of NaCl negatively affected all measured parameters. In contrast, gibberellic acid applied at 200 ppm was effective on most of the parameters measured, particularly under 100 mM of NaCl. These findings indicate that immersing seeds in gibberellic acid could mitigate the adverse impacts of salinity.

Seeds of the plant Eucommia ulmoides Oliver, have prominent dormant characteristics. In the present study, seed dormancy and germination of E. ulmoides were investigated by treating them with exogenous GA3 and different water temperatures. The results revealed that exogenous GA3 and warm water soaking treatments were beneficial to the dormancy breaking and germination of seeds. The germination rate of seeds treated with 50℃ water was 84.67%. In comparison, the seeds treated with 300 mg·L-1 GA3 had only a germination rate of 45%. This low germination rate resulted probably due to the low temperature of the GA3 solution. Another important reason is the presence of eucommia gum in the seed coat of E. ulmoides which hinders the germination process. During germination, the SOD activity in the seeds treated with GA3 (300 mg·L-1) and warm water (50 °C) increased significantly. Increased SOD activity reduced the degree of oxidative damage of the plasma membrane and resulted a continuous decrease in TBARS content. Thereby, the seeds were prompted to develop in a direction conducive to germination. In addition, GA3 (300 mg·L-1) and warm water (50℃) treatments increased the content of endogenous GA3, IAA, and ZR in the seeds, and decreased the content of endogenous ABA. This had resulted significantly higher ratios of GA3/ABA, IAA/ABA and ZR/ABA than those ratios of distilled water (room temperature) treated plants. In short, 50 °C warm water treatment has a more marked germination promoting effect on E. ulmoides seeds.
