Effect of psoralen on the regulation of osteogenic differentiation induced by periodontal stem cell-derived exosomes

Abstract: Periodontitis is a chronic inflammatory disease that is the main cause of tooth loss in adults, and the key to periodontitis treatment is the repair and regenerate of periodontal bone tissue. Psoralen is the main component of the Psoralea corylifolia Linn, which shows antibacterial, anti-inflammatoryand osteogenic activities. It promotes the differentiation of periodontal ligament stem cells toward osteogenesis. Exosomes secreted by stem cells play important roles in information transmission during the osteoge… Show more

“…2 D and F). Furthermore, the effect of 10 μg/mL PSO on the osteogenic ability of PDLSCs has been demonstrated by our previous research [ 38 ]. Therefore, this study determined that the optimal concentration of SDF-1 was 100 ng/mL, and the optimal concentration of PSO was 10 μg/mL.…”

“…Human periodontal ligament stem cells (hPDLSCs) were obtained according to previous studies. 50 hPDLSCs from the periodontal ligament of healthy third molars without any dental and periodontal tissue disease. The third molars used were all from patients (18–25 years old) without systemic diseases.…”

“…The stimulation of periodontal stem cells with psoralen, a compound extracted from Psoralea corylifolia, changed the pattern of miRNAs secreted in exosomes. Vesicles derived from stimulated cells were associated with a reduced expression of miR-125b-5p, which is a molecule that inhibits osteogenesis [161].…”
