Effect of the sesterterpene-type metabolites, ophiobolins A and B, on zygomycetes fungi

Abstract: Ophiobolins are sesterterpene-type phytotoxins produced by fungi belonging mainly to the genus Bipolaris. In this study, the antifungal effect of ophiobolins A and B on different zygomycetes has been examined. Depending on the zygomycete tested, MIC values of 3.175-50 μg mL(-1) were found for ophiobolin A and 25-50 μg mL(-1) for ophiobolin B. Ophiobolin A inhibited sporangiospore germination of Mucor circinelloides and caused morphological changes; the fungus formed degenerated, thick or swollen cells with sep… Show more

“…Fragmentation of nuclei was detected in resting PK-15 cells after the exposure of the monolayer to 6-epi-ophiobolin K-, ophiobolin K-, and ophiobolin G-containing crude extract from A. calidoustus MH34, which has also been described in relation to ophiobolin A in the L1210 cell line. An apoptosis-like cell death process was also observed in M. circinelloides after 1.6 µg mL −1 ophiobolin A treatment and for ophiobolin O in MCF-7 cells [23][24][25][26]. Ophiobolin A was able to induce paraptosis-like cell death in human glioblastoma multiforme cells by decreasing the big/large conductance Ca 2+ -activated K + channel activity [8,9].…”

“…In addition to T-toxin for example, C. heterostrophus also produces ophiobolins that are sesquiterpene phytotoxins ( Fig. Ophiobolins are also active against certain Zygomycetes as they inhibit sporangiospore germination of Mucor circinelloides and in addition cause morphological defects (Krizsan et al, 2010). A mechanism for their antimicrobial and antitumor activity was proposed based on competition for binding to the membrane ivermectin receptor, which modulates an invertebrate specific glutamate-gated chloride channel (Tsipouras et al, 1996).…”

“…These six subclasses of sesterterpenoids have been accounted for to display noteworthy cytotoxicities against tumor cells [28]. Ophiobolins have pulled in across the board consideration due to their phytotoxic, antimicrobial, nematocidal and cytotoxic bioactivities [29,30] (Figure 14-16).…”
