Effect of thermal and chemical modifications on the mechanical and release properties of paracetamol tablet formulations containing corn, cassava and sweet potato starches as filler-binders

Abstract: A B S T R A C TObjective: To investigate the effects of acetylation and pregelatinization of cassava and sweet potato starches on the mechanical and release properties of directly compressed paracetamol tablet formulations in comparison with official corn starch. Methods: The native starches were modified by acetylation and pregelatinization. The tablets were assessed using friability (F r ), crushing strength (C s ), disintegration time (D t ) and dissolution parameters. Results: Starch acetylation produced p… Show more

“…The interest in DC technique draws from its simplicity (very few processing steps), cost‐effectiveness (few materials used), and suitability for materials sensitive to moisture . Direct compression formulation also produces consistent dissolution profiles in tablets . Table shows the processing steps involved in direct compression in comparison with dry and wet granulation methods of tablet manufacture.…”

“…Disintegration time is usually a necessary step for dissolution, and it could be the rate-determining step in the process of drug absorption. 25 This is because a drug entity must be present in solution before it can be absorbed, and this requires an initial disintegration process. HPMC, the longest bioadhesion time occurred with tablet F2.…”

“…Binder efficiency measures the interaction between tablet hardness, friability and disintegration time. A high binder efficiency implies that tablets formulated with it possess excellent hardness, low friability (<< 1%) and short disintegration time (35,36).…”
